Title:Vpliv stopnje zrelosti na celokupno antioksidativno učinkovitost v različnih sortah sliv

Abstract:V okviru diplomske naloge smo določali celokupno antioksidativno učinkovitost (AOU) v različno zrelih plodovih štirih sort sliv 'Čačanska lepotica', 'Valor', 'Stanley' in 'Tophit'. Plodove smo razkoščičili, homogenizirali in ekstrahirali z raztopino aceton/voda (70 : 30, v/v). V ekstraktih smo določili celokupno AOU z ABTS metodo. Meritve so temeljile na redukciji ABTS·+ radikala, ki se tvori pri oksidaciji med ABTS (2,2'azino-bis (3-etilbenztiazolin 6-sulforonska kislina)) in kalijevim peroksodisulfatom in ga določimo spektrofotometrično pri valovni dolžini 734 nm. Umeritvena krivulja je bila v izbranem koncentracijskem območju linearna s korelacijskim koeficientom večjim od 0.99. AOU je bila odvisna od sorte in od stopnje zrelosti. Med zorenjem se je kazal trend naraščanja AOU do stopnje optimalne zrelosti. V optimalno zrelih plodovih je bila najvišja AOU ocenjena pri sorti 'Valor' (18.6 µmol Trolox ekvivalenta (TE)/g svežega vzorca (SV)), sledili sta sorti 'Čačanska lepotica' (12.8 µmol TE/g SV) in 'Stanley' (11.4 µmol TE/g SV), najnižja vrednost pa je bila določena pri sorti 'Tophit' (10.8 µmol TE/g SV). Glede na to, da so bile vse sorte vzorčene na isti lokaciji in pridelane na podoben način, lahko razlike v AOU pripišemo genetski variabilnosti in z njo povezanimi razlikami v biosintezi fenolnih sekundarnih metabolitov.

Title:The effect of ripening stage on the total antioxidant activity of selected plum cultivars
Abstract:The aim of the graduation thesis was to determine the total antioxidant activity (TAA) in fruits of four different plum cultivars 'Čačanska lepotica', 'Valor', 'Stanley' and 'Tophit' during the ripening period. Plums were pitted, homogenised and extracted with acetone/water (70 : 30, v/v). TAA in plum extracts were determined by ABTS method. Measurements were based on the reduction of radical cation (ABTS•+) which was formed through the reaction of oxidation between ABTS (2,2΄-azino-bis-(3-ethylbenzothiazolin-6-sulfonic acid)) and potassium peroxodisulphate. The remaining amount of ABTS•+ was determined spectrophotometrically at wavelength of 734 nm. Calibration curve was linear in the chosen concentration range with a correlation coefficient higher than 0.99. Antioxidant activity depended on cultivar and ripening stage. Ripening resulted in an increased TAA in plums. When plums reached the optimal eating quality, the highest antioxidant activity was estimated in ‘Valor’ plums (18.6 μmol of Trolox equivalent (TE)/g of fresh weight (FW)), followed by cultivars 'Čačanska lepotica' (12.8 μmol TE/g FW) and 'Stanley' (11.4 μmol TE/g FW), while cultivar 'Tophit' showed the lowest antioxidant activity (10.8 μmol TE/g FW). Regarding the fact that plum cultivars were grown in the same location under the same horticultural practices, the differences in TAA could be ascribed to genetic variability affecting the differences in biosynthesis of phenolic secondary metabolites.
