Title:RAZMNOŽEVANJE PODLAGE ČEŠNJE GISELA 5 V IN VITRO RAZMERAH (Prunus cerasus × P.canescens)

Abstract:V obdobju od junija 2008 do julija 2010 smo na Fakulteti za kmetijstvo in biosistemske vede v Mariboru opravili poskus, v katerem smo proučevali možnosti razmnoževanja podlage Gisela 5 v tkivni kulturi. V poskus mikropropagacije smo vključili 44 brstov Gisela 5, ki smo jih inokulirali na indukcijsko gojišče. Brste smo predhodno sterilizirali po dveh metodah. Z dikloroizocianurno kislino smo sterilizirali 23 brstov, od katerih jih je preživelo 95,5 %. Po drugi metodi, kjer smo 21 brstov sterilizirali z natrijevim hipokloritom, je preživelo 57,1 % brstov. Na gojišči G1 in G2 smo subkultivirali 80 vitalnih poganjkov ter opazovali njihovo rast. Iz 40 poganjkov smo na G1 namnožili 119 poganjkov, na gojišču G2 pa 169. Gojišče G2 se je izkazalo za boljše, saj je bil tukaj odstotek kalusiranih in propadlih poganjkov nižji kot pri G1. Dovolj razvite rastline smo prestavili na štiri gojišča za koreninjenje (K1, K2, K3, K4), ki so se razlikovala po vsebnosti avksinov (IBA in NAA). Največ rastlin je koreninilo na gojišču K3 z 0,5 mg/l IBA (90 %), najmanj (65 %) pa na gojišču K4 z 1 mg/l NAA. Ukoreninjene rastline smo presadili v substrat in jih aklimatizirali v rastni komori v laboratoriju oz. meglilni komori rastlinjaka. Aklimatizacija v meglilni komori je bila v primerjavi z rastno komoro boljša, saj je aklimatizacijo v meglilni komori preživelo več rastlin (36,4 %) kot v rastni komori (11,5 %).

Title:IN VITRO PROPAGATION OF CHERRY ROOTSTOCK GISELA 5 (Prunus cerasus × P.canescens)
Abstract:The experiment was carried out from June 2008 until July 2010 at the Faculty of Agriculture and Life Sciences of Maribor. The main purpose of this study was to obtain the best protocol for micropropagation of Gisela 5 in vitro. The micropropagation experiment included 44 Gisela 5 buds that were inoculated onto induction culture medium. The buds were previously sterilized by two methods. The method of dichloroisocyanuric acid was used for sterilized 23 buds, of which 95.5% survived. Based on another method, 21 buds where sterilized by using sodium hypochlorite, 57.1% of buds survived. 80 vital sprouts were subcultivated on cultural mediums G1 and G2 and observed their growth. From 40 sprouts 119 sprouts were reproduce on the G1 medium, and 169 on the G2 medium. The G2 culture medium has proved to be better, since the percentage of callused and failed sprouts was lower than in the case of G1 culture medium. Sufficiently developed plants were moved to four culture media for rooting (K1, K2, K3, K4), which differed in the content of auxines (IBA and NAA). In the culture medium K3 with 0.5 mg/l IBA the percentage of rooted plants was the highest (90%) compared with medium K4 with 1mg/l NAA, where the lowest percentage (65%) of rooted plants was obtained. Rooted plants were transplanted into the substrate and were acclimatized in the growth chamber in the laboratory or fog-like chamber of the greenhouse. Acclimatization in the fog-like chamber was better compared to the growth chamber, since acclimatization in the fog-like chamber survived more plants (36.4%) compared to the growth one (11.5%).
