Title:RAZMNOŽEVANJE VRTNIC (Rosa spp.) V IN VITRO RAZMERAH

Abstract:Uspešno razmnoževanje v in vitro razmerah je odvisno od materinih rastlin, sterilizacije, sestave gojišč, natančnosti, pogojev gojenja in aklimatizacije. V obdobju od oktobra 2009 do decembra 2010 smo na Fakulteti za kmetijstvo in biosistemske vede v Mariboru opravili poskus, v katerem smo proučevali možnosti razmnoževanja vrtnic (Rosa spp.) v tkivni kulturi. V poskus mikropropagacije smo vključili 5 genotipov vrtnic (G1, G2, G3, G4 in G5). Z dikloroizocianurno kislino (sterilizacija S1) smo sterilizirali 25 brstov G1, 34 brstov G2, 93 brstov G3 in 23 brstov G5. S sterilizacijo na osnovi natrijevega hipoklorita (sterilizacija S2) pa smo sterilizirali 25 brstov G1, 16 brstov G2, 88 brstov G3, 80 brstov G4 in 24 brstov G5. Po sterilizaciji 1 je bilo med 175 nastavljenimi brsti 64 okuženih (36,6 %), 54 jih je propadlo (30,9 %), 57 (32,6%) pa preživelo. Pri sterilizaciji 2 smo skupaj nastavili 233 brstov, od katerih je 48 (20,6%) propadlo, 168 (72,1 %) jih je bilo okuženih, preživelo pa je 17 (7,3 %) brstov. Sterilizacija 1 je bila boljša v primerjavi s sterilizacijo 2, saj smo s to metodo uspeli dobiti večji delež (32,6 %) vitalnih rastlin v primerjavi s sterilizacijo 2 (7,3 %). Preživele sterilne poganjke smo prenesli na regeneracijsko gojišče. Uspešno smo namnožili genotip 2, kjer smo iz 16 - ih nastavljenih brstov dobili 186 rastlin v 7 mesecih, genotip 3, kjer smo iz 26 - ih nastavljenih brstov dobili 19 rastlin v 4 mesecih in genotip 5, kjer smo iz 30 brstov vzgojili 88 rastlin v 4 mesecih. Dovolj razvite rastline smo prestavili na gojišča za koreninjenje, ki so se razlikovala po vsebnosti avksina (IBA), saharoze in anorganskih soli (MS). Največ (82 %) rastlin G2 je koreninilo na gojišču K3, ki je bilo brez dodanega avksina IBA in je vsebovalo 1/2 MS in 20 g/l saharoze, najmanj (71,4 %) pa na gojišču K2, ki je bilo prav tako brez hormonov in je vsebovalo 1/4 MS ter 30 g/l saharoze. Rastline G3 so najbolje koreninile na gojišču K1 z 0,5 mg/l IBA, 1 MS in 30 g/l saharoze (55,6 %), najslabše pa na gojišču K2 (40 %). Genotip 5 je najbolje koreninil na gojišču K2 (65,9 %), najslabše pa na gojišču K1(38,4 %). Ukoreninjene rastline smo presadili v substrat in jih aklimatizirali v rastni komori v laboratoriju oz. v meglilni komori v rastlinjaku. Aklimatizacija v meglilni komori je bila boljša v primerjavi z rastno komoro, saj je aklimatizacijo v meglilni komori preživelo več rastlin (90,9 %) v primerjavi z rastno komoro (68,1 %).

Title:In vitro propagation of rose (Rosa spp.)
Abstract:A successful reproduction in in vitro conditions depends on the parent plant, sterilization, media composition, precision, conditions of cultivation and acclimatization. Between October 2009 and December 2010 an experiment was conducted at the Faculty of Agriculture and Life Sciences of Maribor where the ability of rose (Rosa spp.) reproduction in tissue cultures was studied. In the process of micro propagation, 5 rose genotypes (G1, G2, G3, G4, G5) were included. 25 G1 buds, 35 G2 buds, 93 G3 buds and 23 G5 buds were sterilized with dichloroisocyanuric acid (sterilization S1). With sterilization (S2) based on sodium hypochlorite, 25 G1 buds, 16 G2 buds, 88 G3 buds, 80 G4 buds and 24 G5 buds were sterilized. After sterilization S1, there were 64 infected (36.6 %), 54 died (30.9%) and 57 survived (32.6%) out of 175 buds. 233 buds were used for sterilization S2, where 48 (20.6 %) died, 168 (72.1 %) were infected but only 17 (7.3 %) survived. Sterilization S1 was better (32.6 % of vital buds) in comparison to sterilization S2 (7.3 % of vital buds). The sterilized buds were transferred onto a regeneration nutrient medium. Genotypes G1, G3 and G5 were successfully propagated. From G1 genotype186 plants in 7 months were obtained, from G219 plants in 4 months were obtained and from G5 88 plants in 4 months were reproduced. Sufficiently developed plants were moved onto rooting nutrient mediums, which differed in auxin content (IBA), saccharose and anorganic salts (MS). In genotype G2, the biggest share of rooting plants (82 %) was in the K3 nutrient medium without auxin IBA added and contained ¼ MS and 30g/l of saccharose. The most successful rooting of G3 plants was on K1 with 0, 5 mg/l IBA, 1 MS and 30g/l of saccharose (55.6%) while the least successful rooting was on K2 (40%). Genotype 5 rooted most successfully on K2 (65.9%) and least on K1 (38.4%). Rooted plants were transplanted into a substrate and then acclimatized in the laboratory growth chamber or in the greenhouse chamber. Greenhouse chamber acclimatization was better with 90.9% of surviving plants in comparison to the growth chamber with 68.1% of surviving plants.
