Title:Določanje fotosinteznih pigmentov v listih verben po spektrofotometrični in HPLC metodi

Abstract:V prvem sklopu diplomskega dela smo pri določevanju fotosinteznih pigmentov ocenili primerljivost rezultatov dobljenih s spektrofotometrično metodo in tekočinsko kromatografijo (HPLC). S spektrofotometrično metodo smo določili višje vsebnosti fotosinteznih pigmentov. V drugem sklopu poskusa smo preverjali vpliv tretiranja z izvlečkom rjavih alg Ecklonia maxima Kelpak (Kelp products) na vsebnost fotosinteznih pigmentov v potaknjencih verben (Verbena hybrida 'Ipanema blue'). Namen diplomske naloge je bil spremljati dinamiko odziva pigmentov na tretiranje s Kelpakom. Rastline smo v časovnem razmiku treh tednov dvakrat talno tretirali z vodno raztopino Kelpaka (0 %, 0,5 %, 1 % in 2 %). V drugem terminu vzorčenja so imele rastline tretirane z 1 % vodno raztopino Kelpaka, v primerjavi s kontrolo, 14 % nižjo vsebnost klorofila a in 25 % nižjo vsebnost karotenoidov. Na vsebnost fotosinteznih barvil v potaknjencih verben je najugodnejše vplivalo drugo tretiranje s Kelpakom (tretji termin vzorčenja). Vsebnost vseh obravnavanih fotosinteznih barvil je, v primerjavi s kontrolo, v tem terminu najbolj narasla pri tretiranju z 1 % in 2 % vodno raztopino Kelpaka. V zadnjem vzorčnem terminu smo povišano vsebnost fotosinteznih pigmentov določili v vseh tretiranih vzorcih. Na podlagi rezultatov poskusa lahko zaključimo, da je imelo tretiranje z 1,0 % in 2,0 % vodno raztopino Kelpaka pozitiven učinek na vsebnost in razmerja posameznih fotosinteznih pigmentov.

Title:Determination of photosynthetic pigments in vervain leaves by spectophotometric and HPLC methods
Abstract:In the first part of this graduation thesis, we compared the results obtained by spectophotometric as well as HPLC method for the determination of photosynthetic pigments. Higher concentrations of photosynthetic pigments were detected using spectrophotometric method. In the second part of the experiment, we studied the effects of the treatment with brown algae extract Ecklonia maxima Kelpak (Kelp products) on the concentration of photosynthetic pigments in leaves of the vervain cuttings (Verbena hybrida 'Ipanema blue'). The purpose of this graduation thesis was to monitor the dynamic changes in pigment contents during the adventitious root formation of cuttings treated with Kelpak solutions (0 %, 0,5 %, 1 % and 2 %) two times during the period of three weeks. At the second sampling period, plants that had been treated with 1 % Kelpak solution had 14 % lower concentration of chlorophyll a and 25 % lower concentration of carotenoids, when compared to those of control plants. At the third sampling period, after the second treatment, we noticed a beneficial effect on the photosynthetic pigment concentrations. Compared to the control plants, concentrations of all photosynthetic pigments were the highest in plants treated with 1 % in 2 % Kelpak solution. At the last sampling period, we noticed higher levels of photosynthetic pigments in all treated plants. Based on the presented study, we can conclude that the treatment with 1,0 % and 2,0 % Kelpak solution had positive effects on the concentrations and ratios of individual photosynthetic pigments.
