Title:Ekstrakcija aktivnih učinkovin iz baldrijana in vednozelenega gornika

Abstract:Zdravilne rastline in njihovi pripravki so najstarejša oblika zdravljenja najrazličnejših bolezni in še danes so dvema tretjinama človeštva zdravilne rastline glavna pomoč pri zdravljenju. Tudi številna sodobna zdravila neposredno ali v delno spremenjeni obliki izvirajo iz rastlinskih izvlečkov. V tej diplomski nalogi smo predstavili ekstrakcijo aktivnih učinkovin iz baldrijana in vednozelenega gornika, ki spadata med dobro raziskani zdravilni rastlini. Baldrijan ali zdravilna špajka (Valeriana officinalis) je eno najbolj znanih zeliščnih pomirjeval. Zaradi valepotriatov in valerenske kisline, ki jih rastlina največ vsebuje v svojih podzemnih delih, deluje kot sredstvo proti manjšim živčnim motnjam in težavam s spanjem. Vednozeleni gornik (Arctosphyllos uva-ursi) vsebuje fenolne glikozide, zlasti arbutin, ki po pretvorbi v hidrokinone delujejo kot zelo učinkovit antibiotik. Gornik pomaga pri zdravljenju okužb in vnetij sečil, kot sta vnetje mehurja in sečevoda, pa tudi pri zastrupitvi s hrano in diareji. S številnimi poizkusi smo določili optimalne pogoje za izvedbo tehnološkega procesa ekstrakcije baldrijana in vednozelenega gornika. Spreminjali smo različne parametre, temperaturo in čas ekstrakcije, koncentracijo vodne raztopine etanola, število stopenj ekstrakcije in kateri proizvajalec nam lahko nudi najboljši material. Vsebnosti aktivnih učinkovin v suhih ekstraktih smo spremljali z uporabo tekočinske kromatogtafije visoke ločljivosti - HPLC. Optimalen proces je bil nato prenesen v industrijsko merilo, pri katerem je bila izvedena validacija treh zaporednih šarž, specifikacije končnih produktih pa so morale ustrezati zahtevam kupca.

Title:Extraction of active compounds from valerian and bearberry
Abstract:Medicinal herbs and their preparations are the oldest form of healing different diseases, and still today two thirds of the human population uses herbs as the main help in the treatment of the disease. Furthermore, numerous modern medications originate from herbal extracts either directly or in partially changed form. In this thesis, we have presented the extraction of active components from valerian and bearberry, which are well researched herbs. Valerian (Valeriana officinalis) is one of the most known herbal sedatives. The herb contains valepotriares and valeric acid in their underground parts, which function as a remedy against minor nerve disturbances and in sleeping problems. Bearberry (Arctosphyllos uva-ursi) contains phenol glycosides, especially arbutin, that converse into hydroquinons and function as a very effective antibiotic. Bearberry is effective in treatment of infections and inflammations of the urinary system, such as cystitis or urethritis, as well as food poisoning and diarrhea. Optimal conditions for the technological process of the extraction of valerian and bearberry have been defined through numerous experiments. We were changing different parameters, such as temperature and extraction time, and researched what share of ethanol is the most efficient, how many levels of extraction is still reasonable and which producer can offer the best material. We used high pressure liquid chromatography (HPLC) for changing the value of active components in dry extracts. Optimal process was later on transferred to production scale and the validation of three consecutive batches was made with premise that specifications of the final products have to response to the demand of our costumers.
