Title:Karakterizacija mikrobne združbe iz šaržnega biološkega reaktorja za odstranjevanje azo barvil in primerjava učinkovitosti mikrobnega sistema s kombiniranim mikrobno-fizikalnim sistemom

Abstract:Namen diplomske naloge je bil, izolirati, opisati in identificirati mikroorganizme iz aktivnega blata za razgradnjo azo barvil v šaržnem bioreaktorju SBR ter ovrednotiti učinkovitost delovanja biosistema in biosistema v kombinaciji z ultrafiltracijo. V ta namen smo vzpostavili bioproces v bioreaktorju SBR, katerega delovanje je sestavljala reakcijska faza brez dovajanja kisika in faza z vnosom kisika. V bioreaktor smo na začetku inokulirali aktivno blato iz aeriranega bazena centralne čistilne naprave, nadalje pa smo ga dnevno dohranjevali s sveže pripravljeno modelno vodo z barvilom, poimenovanim kislo-rumeno-256 (ang.: acid yellow 256) in hranili za mikroorganizme. Med obratovanjem smo spremljali naslednje parametre: kemijsko potrebo po kisiku (KPK), absorbanco, koncentracijo blata in vrednost pH. Biološko očiščen iztok smo dodatno prečistili z ultrafiltracijo. Ko je biološki sistem deloval optimalno, se je vrednost KPK znižala za 91,09 %, odstranjenih pa je bilo 84,79 % barvil, v povprečju pa je sistem znižal KPK za 73,57 % in odstranil 71,80 % barvil. Po ultrafiltraciji sta se učinkovitost odstranjevanja KPK in razbarvanja zvišali v povprečju za 32,01 % in 61,80 %. Po vzpostavitvi ustreznih pogojev za odstranjevanje barvil in KPK, in prilagoditvi mikroorganizmov, smo iz reaktorja vzeli vzorec aktivnega blata, iz njega izolirali mikroorganizme v aerobnih razmerah gojenja na kompleksnem hranilnem trdnem gojišču z dodatkom azo barvil, ter izolatom preiskali nukleotidna zaporedja odsekov genov za 16S rRNA. Primerjava nukleotidnih sekvenc s sekvencami homolognih genov dostopnih v bazi EMBL/GenBank/DDBJ, je izolate identificirala kot naslednje bakterije: Serratia marcescens, Klebsiella oxytoca, Morganella sp., Elizabethkingia sp., Comamonas testosteroni in Serratia sp.

Title:Characterization of the microbial community from the sequencing batch reactor for azo dyes removal and the efficiency comparison of the microbial system with combined microbial-physical system
Abstract:The goal of this diploma work was to isolate and identify the microorganisms from activated sludge of sequencing batch reactor (SBR) for azo dyes removal and to evaluate efficiency of the biological system and biological system in a combination with ultrafiltration. The SBR process consisted of a reaction step without oxygen followed by the aerated reaction phase. The aerobic activated sludge from the local sewage wastewater treatment plant was used as a seed. The reactor was daily fed with freshly prepared synthetic wastewater containing azo dye acid-yellow-256, named after colour index, and nutrients for microorganisms. The following parameters were monitored throughout the operation period of SBR: chemical oxygen demand (COD), absorbance, sludge concentration and pH. Biologically treated effluent was further purified with ultrafiltration. The efficiency of biological treatment as judged by COD and colour removal was 73.57% and 71.80% in average, respectively and 91.09% and 84.79%, respectively, when SBR was operated optimally. After ultrafiltration the efficiency of the process increased in average for 32.01% and 61.80%, respectively, as judged by COD and colour removal. When the adequate conditions for COD and dye removal were established and after the adaptation of microorganisms to these conditions, the samples of activated sludge were collected for microbial characterization. Bacterial strains were isolated on nutrient solid media containing azo dye, under aerobic conditions, and identified by sequence analysis of a part of the genes coding for 16S rRNA. The following bacterial genera and species were identified: Serratia marcescens, Klebsiella oxytoca, Morganella sp., Elizabethkingia sp., Comamonas testosterone and Serratia sp.
