Title:RAZMNOŽEVANJE BEZGA V IN VITRO RAZMERAH

Abstract:V obdobju od junija 2008 do junija 2010 smo na Fakulteti za kmetijstvo in biosistemske vede opravili poskus, v katerem smo preučevali možnost razmnoževanja različnih genotipov bezga (Sambucus nigra, S. ebulus, S. javanica × S. ebulus, S. javanica × S. nigra) v in vitro pogojih. Preizkusili smo tri načine sterilizacije: z dikloroizocianurno kislino (15 minut in 20 minut) in natrijevim hipokloritom (20 minut). Kot najbolj uspešna se je pokazala sterilizacija z dikloroizocianurno kislino (15 minut). Sterilne in vitalne poganjke smo inokulirali na tri različna gojišča (G1, G2 in G3) za namnoževanje. Na posameznih gojiščih smo poganjke namnoževali 5– 6 tednov in izračunali množitveni faktor. Največji množitveni faktor smo dobili pri genotipu S. ebulus na gojišču G3, in sicer 3,9. Za koreninjenje smo uporabili tri različna gojišča (K1, K2 in K3) in ugotovili, da so se poganjki ukoreninili samo na gojišču K1. Na gojišče K1 smo inokulirali 30 poganjkov S. ebulus, 25 poganjkov S. javanica × S. ebulus, 20 poganjkov S. nigrain 25 poganjkov S. javanica × S. nigra. Ugotovili smo, da S. javanica × S. nigra in S.nigra na gojišču K1 nista koreninila, zelo dobro sta koreninila S. javanica × S. ebulus (100 %), nekoliko slabše pa S.ebulus (76,7 %). Ukoreninjene poganjke smo aklimatizirali v rastlinjaku, v meglilni komori. Aklimatizacija je bila 100 % uspešna.

Title:IN VITRO PROPAGATION OF ELDERBERRIES
Abstract:The investiigation took place from June 2008 to June 2010, at the Faculty of Agriculture and Life Sciences in Hoče near Maribor. We studied the possibility of in vitro propagation in different elderberry genotypes (Sambucus nigra, S. ebulus, S. javanica × S. ebulus, S. javanica × S. nigra). We tested three different substance of sterilization: sodium dichloroisocyanurate – DICA (15 and 20 minutes) and sodium hypochlorite (20 minutes). Sterilization based on sodium dichloroisocyanurate for fifteen minutes proved to be the best suitable. Sterile and vital shoots were inoculated in three different mediums (G1, G2 and G3). We multiplied different shoots in different mediums for 5-6 weeks and calculated the multiplicatiopn factor. The value (3.9) was obtaind for S. ebulus in medium G3. Three different mediums (K1, K2 and K3) were used for rooting. The shoots rooted only in medium K1. We inoculated 30 S. ebulus shoots, 25 S. javanica × S. ebulus shoots, 20 S. nigra shoots and 25 S. javanica × S. nigra shoots in medium K1. We found out that S. javanica × S. nigra and S. nigra on medium K1 did not develop root. The hybrid S. javanica × S. ebulus rooted very well (100 %), while the level of rooting of S. ebulus was a bit lower (76.7 %). The rooted shoots were acclimated in a mist- blow chamber in the greenhouse. The success of the acclimatization was 100 %.
