Title:Značilnosti listov pogostih spomladanskih rastlin v podrasti bukovega gozda

Abstract:Zgodnje pomladanske rastline cvetijo v listopadnih gozdovih še preden zeleni listi v krošnjah dreves zastrejo svetlobo rastlinam v podrasti. Poskušala sem prepoznati skupine spomladanskih rastlin glede na njihove anatomske in morfološke značilnosti listov. Proučevala sem 13 izbranih rastlinskih vrst spomladanske podrasti v ilirsko kolinskem nevtrofilnem bukovem gozdu. Terensko delo v sklopu raziskave je potekalo v aprilu 2013, v starem, nemotenem, listopadnem gozdu na Mariborskem otoku, kjer uspeva nekaj še posebej pomembnih ilirskih vrst že izven svojega strnjenega areala, t.i. Hayekove vegetacijske črte. Na terenu sem nabrala 15 naključno izbranih popolnoma razvitih, zdravih in nepoškodovanih listov in jih sveže uporabila za študij morfologije ter 10 takšnih za študij anatomije, katere sem po prihodu v laboratorij dala v fiksativ FAA, da so se bolje ohranili. V Laboratoriju za molekularno biologijo na Oddelku za biologijo, na Fakulteti za naravoslovje in matematiko Univerze v Mariboru sem v aprilu 2013 opravila morfološko ter v maju in juniju tega leta anatomsko analizo listov. Pripravila sem sveže mokre mikroskopske preparate. Naredila sem prečni prerez lista, za vsakega od desetih listov po en rez, pri čemer sem izmerila debelino lista, debelini palisadnega in gobastega tkiva ter izračunala razmerje med njima. Hkrati sem pripravila sveže mokre mikroskopske preparate spodnje in zgornje povrhnjice ter preštela listne reže in epidermalne celice na površinsko enoto. Za analizo rastlin na makroskopskem nivoju sem stehtala svežo in suho maso ter določila vsebnost suhe snovi listov – LDMC, izračunala površino ter določila specifično listno površino – SLA, na koncu pa izmerila še maksimalno dolžino in širino vsakega lista. Nazadnje sem naredila še klastrsko analizo z namenom prepoznati skupine spomladanskih rastlin, ki so si podobne glede na značilnosti listov. Uporabila sem pristopa klasične klastrske analize in postopka K-mean clustering ter naredila primerjavo med njima. Pridobljeni rezultati obeh metod klastrske analize nakazujejo, da je optimalno v raziskavo hkrati vključiti anatomske in morfološke značilnosti listov, saj imajo na tak način oblikovane skupine višjo razlagalno moč.

Title:Leaf characteristics of widespread spring understorey plants in beech forest
Abstract:Early spring perennials, bloom in a deciduous forest, before the green leaves in the tree canopy intercept the sun-light available for the understorey plants. I tried to identify groups of spring perennials according to their anatomical and morphological leaf characteristics. I studied 13 selected spring perennials in the understorey of an Illyrian low-land neutrophilic beech forest. Fieldwork was conducted in April of 2013 in the old, undisturbed, ancient deciduous forest on the island Mariborski otok. There some particularly important Illyrian species can be found and their occurrence on the island is regarded as outside of their continuous distribution area, outside the so-called Hayek’s vegetation line. In the field I have gathered 15 randomly selected fully developed, healthy and intact leaves and used them fresh to study the morphology and 10 leaves in the FAA fixative to study their anatomy. In the Laboratory of Molecular Biology at the Department of Biology at the Faculty of Natural Sciences and Mathematics of University of Maribor, I conducted the morphological analysis of leaves in April 2013 and an anatomical analysis in May and June of the same year. I made fresh wet microscope slides. I performed the cross sections for measuring the thickness of the leaf, thickness of the palisade and spongy mesophyll and to obtain data for calculated ratio between them. At the same time, I prepared fresh wet slides of upper and lower epidermis and counted stomata and epidermal cells per surface unit. For the analysis of the plants on a morphological level, I weighed fresh and dry mass, and determined the content of dry matter in leaves - LDMC, calculated the surface area and defined the specific leaf area - SLA, and at the end I measured the maximum length and width of each leaf. Lastly, I used a cluster analysis (classical and K-mean clustering) to identify groups of spring perennials with similar leaf characteristics and performed a comparison between analyses. Obtained clusters have shown the best biological argumentation when both – anatomical and morphological leaf characteristics were included in the analysis. Low explanatory power of clusters was shown for cluster performed from only anatomical or only morphological characteristics.
