Title:Vpliv ekstrakcijskega medija na kvaliteto ekstrakta iz gobe Hericium erinaceus

Abstract:Ekstrakcijski postopki so že od nekdaj učinkoviti za industrijo, predvsem živilsko, saj omogočajo relativno hitro in učinkovito izolacijo bioaktivnih komponent. Vedno večjo pozornost v svetu pridobivajo tudi različna prehranska dopolnila, zato smo si izbrali atraktivno in okusno medicinsko gobo Hericium erinaceus. S pomočjo konvencionalne oziroma klasične ekstrakcije in ultrazvočne ekstrakcije, ki smo ju izvajali pri različnih temperaturah, smo pridobili različne ekstrakte omenjene gobe. Uporabljali smo topila etanol in metanol ter ugotavljali, kako izbira topila in temperatura obratovanja vplivata na kvaliteto ekstrakta. Izvedli smo tudi ekstrakcijo polisaharidov, kjer smo kot ekstrakcijski medij uporabili vodo. Sledila je analiza vzorcev s spektrofotometrijo, kjer smo določali vsebnost totalnih fenolov ter antioksidativno aktivnost, izraženo kot procent inhibicije. Ugotovili smo, da na masni izkoristek in izolacijo biološko aktivnih komponent poleg gostote topila vplivata tudi polarnost in temperatura topila, pri kateri izvajamo ekstrakcijo. Bolj polarno je topilo, večja je vsebnost totalnih fenolov ter večji je procent inhibicije.

Title:Effect of extraction media on the quality of extract from Hericium erinaceus
Abstract:Extraction procedures have always been effective in the industry, especially in agriculture as they allow fast and effective isolation of bioactive components. Globally there is a lot of attention on various food supplements, that is why we have chosen the attractive and tasty medical mushroom Hericium erinaceus. We used conventional extraction procedures as well as more modern methods such as ultrasound extraction. This process involves exposing the samples to different temperatures, therefore gained various different extracts of the aforementioned mushroom. Using ethanol and methanol we tried to asses how solvents as well as temperatures affect the quality of the extracts. We have also carried out extraction of polysaccharides, by using water. In the following step, the samples have been analysed using spectrophotometric methods for determination of total phenols and the total antioxidant assay expressed as a percentage of inhibition. . Mass yield and the isolation rate of biologically active components is mostly affected by the polarity and density of the applied solvent and temperature of the solvent, which also influences solvent density, used in the extraction process. If a solvent with a higher polarity is used, greater phenol content and higher percentage of inhibition is observed.
