Title:Priprava novih piščančjih celičnih kultur

Abstract:Ozaveščenost ljudi o zmanjšanju uporabe živalskih modelov in upoštevanje pravila 3R (reduce, refine, replace = zmanjšaj, dodelaj, zamenjaj) v raziskovalnem svetu je vse večja. Med enega od načinov alternativnih metod sodi tudi delo na celičnih kulturah. Področje razvoja in dostopnosti piščančjih celičnih kultur je, v primerjavi s humanimi in mišjimi, zelo pomanjkljivo. V naši magistrski nalogi smo se odločili razviti nove piščančje celične linije iz primarne kulture pridobljene iz 19 dni starega zarodka piščanca in optimizirati njihovo gojenje in vitro. Najuspešnejša rast celičnih kultur se je z našim protokolom izkazala pri jetrnih hepatocitih ter epiteliju ileuma, nismo pa uspeli vzpodbuditi celic epitelija kolona k delitvi. Te kulture smo v in vitro pogojih s precepljanjem obdržali 238 dni, dodatno pa smo iz njih uspeli klonirati in pripraviti tri celične linije hepatocitov in eno linijo epitelija ileuma. Glede na celično obliko smo primerjali pripravljene kulture celic s komercialno dostopnimi celičnimi kulturami in ugotovili skladnost morfologije. V prihodnosti bi bilo potrebno linije dodatno okarakterizirati na prisotnost značilnih celičnih markerjev in optimizirati pogoje za rast kolonocitov.

Title:Development of new chicken cell culture lines
Abstract:Awareness of people about reducing the use of animal models and compliance with 3R rule (reduce, refine, replace) in the research world is growing. One of the alternative ways is to work with cell culture. The area of development and accessibility of chicken cell cultures is very deficient in comparison with human and mouse cell models. In our master's thesis, we have decided to develop new chicken cell lines from the primary culture derived from the 19-day old chicken embryo and optimize their cultivation in vitro. A successful growth of cell cultures with our protocol was obtained for hepatocellular hepatocytes and the ileum epithelium, but we did not succeed in stimulating growth and division of colon epithelial cells. These cultures were retained 238 days in in vitro conditions. In addition, we managed to clone them and prepare three cell lines of hepatocytes and one cell line of the ileum epithelium. The morphology of newly prepared cell cultures was found similar to commercially available cell cultures. In the future, cell lines should be further characterized for the presence of typical cell markers and the conditions for colonocyte growth optimized.
