Title:Vzpostavitev tkivne kulture pri rastlini noni (morinda citrifolia l.)

Abstract:V letu 2018 smo na Fakulteti za kmetijstvo in biosistemske vede v Mariboru izvedli raziskavo, pri kateri smo ugotavljali uspešnost vzpostavitve tkivne kulture pri rastlini noni (Morinda citrifolia L.). Preizkusili smo dva načina sterilizacije. Pri obeh načinih smo izsečke pred začetkom sterilizacije za 40 sekund tretirali s 70-odstotnim etanolom. Pri prvem načinu sterilizacije smo za sterilizacijo uporabili dikloroizocianurno kislino (DICA), s katero smo izsečke tretirali 8 minut. Pri drugem načinu smo kot sterilizacijsko sredstvo uporabili natrijev hipoklorit (NaClO), s katerim smo izsečke tretirali 8 minut. Sterilizirane izsečke smo inokulirali na gojišče za indukcijo. Kot najuspešnejša se je izkazala osem minutna sterilizacija z DICA, kjer je 100 % rastlin ostalo po sterilizaciji vitalnih in neokuženih. Pri sterilizaciji z natrijevim hipokloritom je 65 % izsečkov propadlo, 16 % jih je bilo okuženih, vitalnih in neokuženih je bilo 19 % in so se okužili (16 %) pri osem minutni sterilizaciji z NaClO. V kasnejših fazah razvoja so se začele pojavljati bakterijske okužbe, zato smo izsečke prestavili na gojišče z dodanim antibiotikom (Cefotaxime sodium), ki ni preprečil pojava okužb.

Title:Establishment of morinda citrifolia l. tissue culture
Abstract:In the investigation, which was carried out at the Faculty of Agriculture and Life Science of the University of Maribor, we investigated the success of establishing Indian mulberry (noni, Morinda citrifolia L.) tissue culture. We used 2 different types of sterilizations of explants. Before sterilization, we treated the explants with 70% ethanol for 40 seconds. The first type of sterilization was based on a treatment with DICA for 8 minutes, whereas the second type was based on a treatment with NaClO for 8 minutes. The sterilized explants were inoculated on an induction medium. The most successful was the 8 minute sterilization with DICA, where all explants remained vital after sterilization. The 8 minutes long treatment with NaClO resulted in 65% deceased and 16% infected explants. In later stages of development, infections began to appear on explants treated with DICA. We transferred some explants to the medium with the antibiotic substance (Cefotaxime sodium) and discovered that this antibiotic was not effective against infections.
