Title:Priprava ekstraktov aronije Aronia melanocarpa in testiranje njihove biološke aktivnosti

Abstract:V magistrskem delu smo pripravili ekstrakte aronije iz liofoliziranih plodov aronije. Ekstrakti so bili pripravljeni s superkritično in s konvencionalno ekstrakcijo. Superkritično ekstrakcijo s topilom CO2 in kotopilom etanolom smo izvedli pri pogojih: 100, 200 in 300 barih ter temperaturah 40 oz. 60 C. Konvencionalna ekstrakcija je obsegala ekstrakcijo s Soxhletovim aparatom, ultrazvočno in hladno ekstrakcijo. Uporabili smo sledeča topila: absolutni etanol, metanol, voda in 50 % etanol. Pripravljenim ekstraktom smo določili vsebnost celokupnih fenolnih spojin in proantocianidinov. Prav tako smo preverili antioksidativne lastnosti pripravljenih ekstraktov. Glede na vsebnost fenolnih spojin smo izbrali nekaj ekstraktov, za katere smo predpostavili, da bi lahko imeli pozitiven učinek na humane intestinalne epitelijske celice. Izbrali smo vse konvencionalne ekstrakte, pripravljene s 50 % etanolom in superkritični ekstrakt, pripravljen pri pogojih 200 barov in 60 C. Izbranim ekstraktom smo določili topnost fenolnih spojin v vodi. Vpliv izbranih ekstraktov aronije na humanih intestinalnih epitelijskih celicah smo določili s testom viabilnosti. Najvišje izkoristke ekstrakcij smo določili pri konvencionalnih ekstrakcijah, kjer so bila uporabljena topila voda in 50 % etanol. Najvišje vsebnosti celokupnih fenolnih spojin in proantocianidinov vsebujejo ekstrakti aronije, pripravljeni s topilom 50 % etanol. Prav tako smo tem ekstraktom določiti tudi najvišje antioksidativne lastnosti. Test viabilnosti je bil uspešen pri vseh treh izbranih konvencionalnih ekstraktih aronije.

Title:Preparation of aronia melanocarpa extracts and testing of their biological activity
Abstract:Extracts of aronia were prepared by supercritical and conventional extraction from lyophilized fruits of aronia. Supercritical extraction with CO2 as solvent and ethanol as cosolvent was carried out at 100, 200 and 300 bars and at temperatures of 40 or 60 °C. Conventional extraction has also been performed and included Soxhlet extraction, ultrasonic and cold extraction. Four different conventional solvents/mixtures have been applied: absolute ethanol, methanol, water and 50% ethanol. The prepared extracts were tested for the content of the total phenolic compounds and proanthocyanidins. Their antioxidative properties have also been examined. Depending on the content of phenolic compounds, extracts assumed to expose a positive effect on human intestinal epithelial cells, have been selected. Those were conventional extracts extracted with 50 % ethanol and supercritical extract, prepared at 200 bar and 60 °C. The solvability of phenolic compounds in water by the selected extracts was determined. The influence of aronia extracts on human intestinal cells was determined with a viability test. The highest extraction yields were determined by conventional extractions, where water solvents and 50% ethanol were used. The highest contents of the total phenolic compounds and proanthocyanidins contain extracts prepared with a solvent of 50% ethanol. These extracts also have the highest antioxidant properties. The viability test was successful in all three selected conventional aronia extracts
