Title:Izolacija in karakterizacija bakterije Clostridum difficile s površine krompirja

Abstract:Clostridium difficile je po Gramu pozitivna, anaerobna, sporogena bakterija, ki povzroča črevesne okužbe, najpogosteje po zdravljenju z antibiotiki, kemoterapiji in po kirurških posegih, ki privedejo do porušenja normalne črevesne mikrobiote. Bakterijo lahko najdemo v različnih naravnih okoljih, črevesju živali in tudi hrani. Namen magistrskega dela je bil izolirati in karakterizirati bakterijo C. difficile s površine krompirjev, kupljenih v različnih lokalnih trgovinah ter iz čim več različnih držav. Iz 8 različnih trgovin smo pridobili 23 vzorcev krompirja, ki so izvirali iz 10 različnih držav. Prisotnost C. difficile smo dokazali pri 14 vzorcih (60,9 %). Iz vseh vzorcev smo osamili 162 izolatov C. difficile, katerim smo najprej s PCR-pomnoževanjem potrdili za C. difficile specifični gen cdd3, ter jih opredelili s PCR-ribotipizacijo in toksinotipizacijo. Vseh 162 izolatov smo uvrstili v 9 različnih PCR-ribotipov: 012, 014/020, 023, 027, 053, 126, 150, SLO 129 in SLO187. Najpogostejši PCR-ribotip je bil 014/020, sledila sta ribotipa 053 in 126. Toksigenih je bilo 137 izolatov, kasneje uvrščenih v 5 različnih toksinotipov – toksinotip 0, I, III, IV in V, 26 izolatov pa je bilo netoksigenih (s PCR nismo mogli dokazati prisotnosti toksinskih genov). Naši rezultati kažejo, da je krompir kontaminiran s sporami bakterije C. difficile v visokem deležu in da lahko na krompirju najdemo enake PCR-ribotipe, kot se pojavljajo pri ljudeh, živalih in v okolju.

Title:Isolation and characterization of Clostridium difficile from potato surface
Abstract:Clostridium difficile is a Gram-positive, anaerobic, sporogenic bacterium that causes intestinal infections, most often after treatment with antibiotics, chemotherapy and after surgery, when normal gut microbiota becomes disbalanced. The aim of the this work was to isolate and characterize C. difficile from the surface of potatoes, purchased in various local supermarkets and originating from different countries. From 8 different stores we acquired 23 samples originating from 10 different countries. From 14 samples (60,9 %) positive for C. difficile we obtained 162 C. difficile isolates. The identity of isolates was confirmed by PCR-amplification of C. difficile specific gene cdd3. PCR-ribotyping and toxinotyping were used to characterize C. difficile isolates. We found 9 different PCR-ribotypes: 012, 014/020, 023, 027, 053, 126, 150, SLO 129 and SLO187, with 014/020 being the most common ribotype, followed by ribotype 053 and 126. A total of 137 isolates were toxigenic and were distributed into 5 different toxinotypes - 0, I, III, IV, and V. For 26 isolates we could not determine presence of toxin genes by PCR. Our results demonstrate high prevalence in potato contamination by C. difficile spores and that same PCR-ribotypes found on potatoes are the same as those isolated from infected humans.
