Title:Izražanje dolge nekodirajoče RNA CCAT1 pri bolnikih z rakom glave in vratu : diplomsko delo univerzitetnega študijskega programa I. stopnje

Abstract:Rak glave in vratu (angl. Head and Neck Cancer, HNC) je heterogena skupina rakov, ki nastanejo v ustni votlini, žrelu, grlu, nosni in obnosni votlini ter na ustnicah, žlezah slinavkah in vratnih bezgavkah. Letno za rakom glave in vratu zboli 600.000 ljudi in umre jih 300.000, kar ga uvršča na sedmo mesto najpogostejših rakov na svetu. Pri razvoju raka glave in vratu sodeluje dolga nekodirajoča RNA (lncRNA) CCAT1 (angl. Colon Cancer Associated Transcript 1), katere vloga in molekulerni mehanizmi delovanja so slabo okarakterizirani. V diplomski nalogi smo na vzorcu 34 bolnikov z RT-PCR proučili izražanje lncRNA CCAT1 v zdravem in tumorskem tkivu pri rakih grla, žrela, ustne votline in ustnic. Izražanje lncRNA CCAT1 se ni razlikovalo med zdravim in tumorskim tkivom in med proučevanimi skupinami rakov. Vpliva stadija in okoljskih dejavnikov tveganja (kajenje, uživanje alkohola, okuženosti s HPV16), na izražanje lncRNA CCAT1 v zdravem in tumorskem tkivu nismo ugotovili. Za rake glave in vratu s perivaskularno, perinevralno in limfovaskularno invazijo je bilo značilno nižje izražanje lncRNA CCAT1 kot za neinvazivne oblike, vendar razlika ni dosegla statistične značilnosti (p=0,08). Naši rezultati kažejo, da kajenje, uživanje alkohola in okuženost s HPV16 ne vplivajo na izražanje lncRNA CCAT1 pri rakih glave in vratu. Za potrditev vloge lncRNA CCAT1 pri patogenezi raka glave in vratu bo potrebno izražanje lncRNA CCAT1 proučiti na večjem vzorcu bolnikov.

Title:Expression of long non-coding RNA CCAT1 in patients with head and neck cancer
Abstract:Head and Neck Cancer (HNC) is a heterogeneous group of cancers that includes cancers of the lips, oral cavity, salivary glands, nasal cavity, paranasal sinuses, pharynx, larynx and head and neck lymph nodes. It is the seventh most common cancer worldwide with 600,000 new cases annually and a mortality rate reaching 300,000 deaths each year. Previous studies have shown increased expression of long noncoding RNA CCAT1 (CCAT1) in HNC and its putative role in cell proliferation, clonality, migration and invasion; however, the exact role and molecular mechanisms of CCAT1 in HNC tumorigenesis is yet to be determined. In the present study, expression of CCAT1 was analysed using RT-PCR on paired tumour-normal tissue samples from 34 patients with HNC. CCAT1 expression did not differ between the tumour and normal tissue and between the tumours of the pharynx, larynx, oral cavity and lips. Similarly, the tumour grade, tobacco smoking, alcohol consumption, HPV16 infection and tumour invasion did not affect CCAT1 expression in HNC. However, there was a statistical trend (p=0.08) towards lower CCAT1 expession in tumours with perivascular, perineural and lymphovascular invasion. Results of the present study are in line with the previous observations suggesting that tobacco smoking, alcohol consumption and HPV16 infection do not affect CCAT1 expression in HNC. Results of the present study require further testing and confirmation on a larger cohort of patients with HNC.
