Title:Vsebnost ščitničnih hormonov in vitamina D3 v krvnem serumu : diplomsko delo univerzitetnega študijskega programa I. stopnje

Abstract:V diplomski nalogi smo določevali vsebnost tiroidnih hormonov, trijodotironina (F3) in tiroksina (T4) ter metabolit vitamina D3 (25-hidroksivitamin D3) v krvnem serumu. Vzorce smo analizirali s pomočjo tekočinske kromatografije v tendemu z masno spektrometrijo. Za določitev vsebnosti ščitničnih hormonov in vitamina D3 v krvnem serumu smo najprej izvedli percipitacijo proteinov. Postopek odstranitve proteinov je bil izveden na dva načina. Ena metoda je proteine iz seruma odstranila s pomočjo centifugiranja, medtem ko je druga metoda obarjala proteine s pomočjo organskega topila. Obe metodi sta dali dobre rezultate, kar pomeni, da ju lahko uporabimo pri načrtovanju eksperimentov. Optimizacijo kromatografskih parametrov smo določili primerno metodo za določevanje tiroidnih hormonov in vitamina D3 v krvnem serumu. Po vzpostavitvi optimalnih pogojev za analizo, smo metodo tudi delno validirali. Dobljeni rezultati validacije so dali zadovoljive rezultate. Metoda je primerna za določevanje trijodotironina v krvnem serumu v območju koncentracije 1,25-250 ng/mL, tiroksina v območju 1,25-250 ng/mL in metabolite vitamina D3 v območju 2,5-250 ng/mL z RSD < 10%. Analizirani vzorci so bili pripravljeni z enostavnim postopkom, ki je časovno ugoden za analizo večjega števila vzorcev. Metoda je dobra alternativa imunološkim metodam, ki zahtevajo veliko časa in so manj natančne.

Title:Content of thyroid hormones and vitamin D3 in blood serum
Abstract:In the diploma thesis we determined the content of thyroid hormones, triiodothyronine (F3) and thyroxine (T4) and metabolized vitamin D3 (25-hydroxyvitamin D3) in the blood serum. Samples were analyzed by trend chromatography by mass spectrometry. To determine the content of thyroid hormones in vitamin D3 in the blood serum, we performed protein precipitation. The protein removal process was performed in two ways. One method removed the protein from the serum by centrifugation, while the other method precipitated the protein by other organs. Both methods provided good results, which means that users can be used in designing experiments. Optimization of chromatographic parameters was precisely determined for the determination of thyroid hormones in vitamin D3 in blood serum. We showed the optimal conditions for analysis, and the method was also partially validated. The selected results are confirmed so that the results are satisfactory. The method is suitable for the determination of triiodothyronine in blood serum at a concentration range 1,25-250 ng/mL, thyroxine in range 1,25-250 ng/mL and metabolite of vitamin D3 in range of 2,5-250 ng/mL with RSD <10%. The analyzed samples were prepared by a simple procedure that was favorable for the analysis of a large number of samples. The method is a good alternative to immunological methods that require a lot of time and so little precision.
