Title:Uporaba različnih izolacijskih postopkov in antioksidativna učinkovitost timijana (thymus vulgaris l.) : diplomsko delo visokošolskega strokovnega študijskega programa I. stopnje

Abstract:Diplomsko delo obravnava različne izolacijske postopke aktivnih komponent iz timijana. Izpeljali smo pet različnih izolacijskih postopkov, ob uporabi različnih topil in pri različnih obratovalnih pogojih (tlakih in temperaturah). Izvedli smo konvencionalne ekstrakcije; ekstrakcijo po Soxhletu, vročo in hladno maceracijo ter ultrazvočno ekstrakcijo ter superkritično ekstrakcijo s CO2 in sotopilom kot naprednejši ekstrakcijski metodi. Uporabili smo dve nepolarni topili, heksan in petrol eter, ter dve polarni topili, etanol in mešanico etanola in vode. Izkoristki ekstrakcij so bili največji pri uporabi polarnih topil etanola in mešanice etanola in vode. Iz tega smo ugotovili, da se v timijanu nahaja več polarnih komponent, kot nepolarnih. Timijan tako vsebuje veliko karvakrola in timola, ki sta nekoliko bolj polarni komponenti, topni predvsem v etanolu. Med nepolarne komponente uvrščamo y-terpinen, p-cimen, a-terpenin, 1,8-cineol. Dobljene ekstrakte smo nato uporabili za nadaljnjo analizo antioksidativne aktivnosti s pomočjo radikalske metode z DPPH. S tem smo ugotovili, da smo največ fenolnih spojin, ki so odgovrne za antioksidativno delovanje izolirali z mešanico topil etanola in vode. Absorbanco referenčne raztopine smo izmerili 0,6348. Vrednosti rezultatov, % inhibicije, pa so bile v območju med 8,02 % in 89,00 %. Prav tako smo skušali določiti vsebnost komponent v eteričnih oljih z plinsko kromatografijo masno spektrometrijo GC/MS. Določili smo vsebnost karvakrola in timola. Vsebnost timola je bila znatno višja od karvakrola. Vsebnost smo podali v µg/g materiala. Vsebnosti timola so bile med 0,534 µg/g materiala in 2,370 µg/g materiala. Vsebnost karvakrola pa med 0,058 µg/g materiala in 0,440 µg/g materiala. Torej naš glavni namen diplomske naloge je bil, da iz timijana izoliramo čim več aktivnih komponent, s pomočjo različnih topil in določimo njihovo antioksidativno aktivnost.

Title:Different isolation methods and antioxidant properties of thyme (thymus vulgaris l.)
Abstract:This thesis deals with different isolation processes of active thyme components. Five different insulation processes were carried out, using different solvents and operating conditions (pressures and temperatures). Conventional extractions were performed; soxhlet extractions, hot and cold maceration and ultrasonic extractions, and supercritical CO2 and solvent extractions as more advanced extractions methods. We used two non-polar solvents, hexane and petroleum ether, and two polar solvents, ethanol and an ethanol/water mixture. Extraction yields were highest using the polar solvents ethanol and ethanol/water mixtures. From this, we found that there are more polar components in thyme than non-polar components. Thyme is also high in carvacrol and thymol, which are slightly more polar components, soluble mainly in ethanol. The non-polar components include y-terpinene, p-cymene, a-terpinene, 1,8-cineole. The isolated extracts were then used for further analysis of antioxidant activity using the DPPH radical method. This showed that most of the phenolic compounds responsible for the antioxidant activity were isolated with a solvent mixture of ethanol and water. The absorbance of the reference solution was 0,6348. The values of the results, % inhibiton, were in the range between 8,02 % and 89,00 %. We also tried to determine the content of the components in the essential oils by GC/MS gas chromatography-mass spectrometry. We determined the content of carvacrol and thymol. Thymol content was significantly higher than carvacrol. The contentn was given in µg/g material. Thymol contents were between 0,534 µg/g of material and 2,370 µg/g of material. The carvacrol content is between 0,058 µg/g of material and 0,440 µg/g of material. So the main aim of our thesis was to isolate as many active components as possible from thyme, using different solvents, and determine their antioxidant activity.
