Title:Izbira metode za določevanje antioksidativne aktivnosti biološko aktivnih komponent : magistrsko delo

Abstract:Antioksidanti so spojine, ki so zmožne inhibirati oksidacijo s čimer ščitijo organizem pred obolenji, povzročenimi s prostimi radikali. V magistrskem delu smo želeli določiti optimalno metodo za določanje antioksidativne aktivnosti biološko aktivnih komponent glede na njihovo strukturo in topnost. Uporabili smo sintetične spojine, ki izkazujejo biološko aktivnost, med katere spadajo askorbinska kislina, butiliran hidroksi toluen, kofein, klorogenska kislina, resveratrol, rutin in teofilin ter ekstrakte iz naravnih surovin, tj. CBD, črna kumina, šipek in zelena kava, ki smo jih pripravili z mletjem in ultrazvočno ekstrakcjio ter uparevanjem z rotacijskem uparjalnikom. Raztopine sintetičnih spojin in ekstraktov smo pripravili v metanolu ter testirali njihove antioksidativne lastnosti z DPPH, CUPRAC, s FRAP in z ORAC testom ter β-karotensko metodo. Dobljene rezultate smo ovrednotili glede na kemijske lastnosti in strukturo spojine ter sklepali, katera izmed antioksidativnih metod je najbolj primerna za določevanje antioksidativne aktivnosti.

Title:Choice of method for determining the antioxidant activity of biologically active components
Abstract:Antioxidants are compounds, capable of inhibiting oxidation, thereby protecting organism from diseases, caused by free radicals. In master's thesis we wanted to determine the optimal method for the determination of the antioxidant activity of biologically active components according to their structure and solubility. We used synthetic compounds exhibiting biological activity, including ascorbic acid, butylated hydroxytoluene, caffeine, chlorogenic acid, resveratrol, rutin and theophylline, as well as extracts from raw materials, i.e. CBD, black cumin, rosehip and green coffee, prepared by grinding, ultrasonic extraction and evaporation with a rotary evaporator. Solutions of synthetic compounds and extracts were prepared in methanol and their antioxidant properties were tested by DPPH, CUPRAC, FRAP, ORAC and β-carotene bleaching assays. Obtained results were evaluated with respect to the chemical properties and structure of the compound and concluded which of the antioxidant assay is the most suitable for determination of antioxidant activity.
