Title:Optimizacija procesa ekstrakcije klorofila iz rastline Alfalfa (Medicago sativa) : magistrsko delo

Abstract:Alfalfa (Medicago Sativa) oz. lucerna je večletna kmetijska rastlina iz družine grahovk in je ena najbolj znanih zdravilnih rastlin. Zaradi vsebnosti beljakovin, vitaminov, mineralov in vlaknin je lucerna zelo hranljiva krma za prežvekovalce. Listi lucerne so dober vir klorofilov, karotenov in antocianov, ki so odgovorni za obarvanost listov v različnih letnih časih. Klorofil, ki ga pridobimo z ekstrakcijo iz rastlin je naravni pigment v oljetopni obliki. Uporaba oljetopnih aplikacij je zaradi slabše stabilnosti omejena, zato se oljetopen klorofil pogosto s postopkom saponifikacije pretvori v vodotopno obliko, imenovano klorofilin, s čimer se njegova uporaba razširi. V magistrskem delu smo optimirali proces ekstrakcije klorofila iz rastline Alfalfa in raziskali postopek pretvorbe oljetopnega magnezijevega klorofila v vodotopno obliko, natrijev bakrov klorofilin. Optimirali smo čas in izbiro topila za ekstrakcijo, ter izbiro topila za saponifikacijo. S spektrofotometrično metodo (UV/VIS) smo določili vsebnost bakra v ekstraktu in izračunali izkoristek ekstrakcije.

Title:Optimization of chlorophyll extraction process from Alfalfa plant (Medicago sativa)
Abstract:Alfalfa (Medicago Sativa) or lucerne, is a perennial agricultural plant in the pea family and one of the best-known medicinal plants. Its protein, vitamin, mineral and fibre content makes alfalfa a highly nutritious feed for ruminants. Alfalfa leaves are a good source of chlorophylls, carotenes and anthocyanins, which are responsible for the colouring of the leaves in different seasons. Chlorophyll obtained by extraction from plants is naturally in an oil-soluble form. The use of oil-soluble applications is limited due to its poor stability, so oil-soluble chlorophyll is often converted via a saponification process into a water-soluble form called chlorophyllin, thus extending its use. In this master thesis, we optimised the extraction process of chlorophyll from the Alfalfa plant and investigated the process of conversion of the oil-soluble magnesium chlorophyll into a water-soluble form, sodium copper chlorophyllin. The time and choice of solvent for extraction and the choice of solvent for saponification were optimised. The copper content of the extract was determined with a spectrophotometric method (UV/VIS) and the extraction yield was calculated.
