Title:Vsebnost flavonoidnih komponent v posušenih olupkih mandarin in njihova antioksidativna aktivnost : magistrsko delo

Abstract:Citrusi, ki pripadajo družini Rutaceae, pomembno prispevajo k zdravju ljudi zlasti zaradi visokih vsebnosti bioaktivnih spojin, med katere prištevamo tudi flavonoide, ki zaradi svojih protimikrobnih, protivnetnih in antioksidativnih učinkov pomagajo pri preprečevanju bolezni. Magistrsko delo prikazuje študijo, namen katere je prikazati potencial olupkov mandarin, ki veljajo za neužitni del ter se običajno zavržejo. Njihova uporaba je zelo široka; od kulinarike, čistil, aromaterapij ter seveda njihove zdravstvene koristi. Uporabljajo se v postopkih ekstrakcije in čiščenja bioaktivnih spojin iz različnih delov plodov, predvsem pri izolaciji karotenoidov, polifenolov, eteričnih olj in barvil. Olupki citrusov zato v tehnološkem in medicinskem pogledu ponujajo vrsto konstruktivnih priložnosti. V magistrskem delu je izvedenih več različnih ekstrakcij (ekstrakcija po Soxhletu, ultrazvočna ekstrakcija in maceracija), s katerimi smo z izbranimi topili ekstrahirali nekatere prisotne flavonoidne komponente iz posušenih olupkov mandarin in jih kvantificirali s kromatografsko analizno metodo LC-MS/MS. Izvedene so bile antioksidativne metode DPPH, FRAP, CUPRAC, ORAC in in vitro metoda simulacije gastrointestinalnega trakta. Rezultati kažejo, da najvišji izkoristek ekstrakcije dobimo pri ultrazvočni ekstrakciji, najnižji pa po metodi Soxhlet. Vsebnost antioksidantov je prav tako najvišja v vzorcih pri ultrazvočni ekstrakciji. Ugotovili smo, da izbira ekstrakcijskega topila vpliva na antioksidativno aktivnost ekstraktov. Vzorci z najvišjo vsebnostjo celokupnih flavnoidov so izkazovali boljše antioksidativne lastnosti. Uporabili smo več testov antioksidativnosti, s katerimi smo dobili delno primerljive rezultate, saj se metode razlikujejo po mehanizmih delovanja.

Title:Content of flavonoid components in dried mandarin peels and their antioxidant activity
Abstract:Citrus fruits, which belong to the Rutaceae family, presents an important impact to human health. Mainly due to the high content of bioactive compounds, including flavonoids, which help prevent diseases, due to their antimicrobial, anti-inflammatory and antioxidant effects. The master's thesis shows a study that aims to show the potential of mandarin peels, which are considered an inedible part and are usually thrown away. Their use is very wide, from culinary, cleaning, aromatherapy, and of course their health benefits. They can also be used in the extraction and purification processes of bioactive compounds from different parts of fruit, such as carotenoids, polyphenols, essential oils and dyes. Therefore, citrus peels offer a range of constructive opportunities in terms of technology and medicine. In the master's thesis, several different extractions (Soxhlet extraction, ultrasonic extraction and maceration) were carried out, with which all the flavonoid components present in dried mandarin peels were extracted with selected solvents and quantified using the LC-MS/MS chromatographic analysis method. Antioxidant methods DPPH, FRAP, CUPRAC, ORAC and in vitro gastrointestinal tract simulation method were performed. The results show that the highest extraction efficiency is obtained with ultrasonic extraction, and the lowest with the Soxhlet method. Also, the content of antioxidants in the samples is the highest with ultrasonic extraction. We found that the choice of extraction solvent affects the antioxidant activity of the extracts. Samples that had a higher content of total flavonoids showed better antioxidant properties. We used several antioxidant tests, with which we obtained partially comparable results, since the methods differ in their mechanisms of action.
