Title:Separacija in karakterizacija vrednih spojin iz zunanje arašidove (Arachis hypogaea L.) lupine z vodo pri podkritičnih pogojih : magistrsko delo

Abstract:Arašid (Arachis hypogaea L.) je strok ali stročnica iz družine Leguminosae. V svetovnem merilu se arašidi pridelujejo predvsem zaradi olja iz semen, so pa tudi pomemben vir beljakovinske hrane v državah v razvoju in v razvitih državah. Lupine arašidov so bogat in poceni stranski proizvodi predelave arašidov. Vsako leto pridelamo kar 5 milijonov ton lupin arašidov. Funkcionalne sestavine v lupinah arašidov so še vedno premalo izkoriščene, zato je gospodarsko zelo pomembno, da se podrobneje razišče lastnosti in možnosti uporabe lupin arašidov. Glavni namen magistrske naloge je bil načrtovanje in optimiziranje postopka izolacije vrednih spojin iz arašidovih olupkov z uporabo metode odzivnih površin z zasnovo Box-Behnkenovega načrtovanja. Za separacijo vrednih spojin smo uporabili okolju prijazno ekstrakcijo s podkritično vodo. Načrt optimizacije na osnovi odzivnih površin nam je omogočil določitev optimalnih pogojev (temperature, časa ekstrakcije in razmerja topilo/material) za maksimiranje izkoristka ekstrakcije in vsebnosti furfuralov v ekstraktih. Pridobljene ekstrakte smo preučevali s pomočjo tekočinske kromatografije visoke ločljivosti (HPLC) ter UV-Vis spektrofotometrije. Določevali smo celokupne ogljikove hidrate, totalne fenole, antioksidativno aktivnost, vsebnost sladkorjev (glukoze in fruktoze) ter derivate sladkorjev (levulinsko kislino, 5-HMF in furfural) Na podlagi rezultatov smo ugotovili, da so optimalni pogoji ekstrakcije za maksimiranje izkoristka in deleža furfurala izoliranega iz arašidovih lupin pri temperaturi 200 °C, času ekstrakcije 45 min in razmerju topilo/material 21 mL/g. Pri teh pogojih je teoretični izkoristek ekstrakcije znašal 30,43 %, eksperimentalni pa 31,36 %. Teoretična učinkovitost izolacije furfurala je bila 33,22 mg/g lupin, eksperimentalno dobljena pa 34,68 mg/g lupin. Pri teh pogojih so pridobljeni ekstrakti pokazali tudi bogat profil biološko aktivnih spojin, vključno s fenolnimi spojinami ter antioksidanti.

Title:Separation and characterisation of valuable compounds from external peanut (Arachis hypogaea L.) shell with water at sub-critical conditions
Abstract:The aim of this master's thesis was the separation of valuable compounds from external peanut (Arachis hypogaea L.) shell using subcritical water. The obtained extracts were characterized, and the response surface methodology with the Box-Behnken design was used for process optimization. The optimization design enabled efficient exploration of the response surface and determination of optimal conditions (temperature, extraction time, and solvent-to-material ratio) for maximizing extraction yield and furfurals content in the extracts. The obtained extracts were analyzed using high-performance liquid chromatography (HPLC) and UV-Vis spectroscopy. Various parameters were determined, including total carbohydrates, total phenols, antioxidant activity, sugar content (glucose and fructose), and sugar derivatives (levulinic acid, 5-HMF, and furfurals). Based on the results, the optimal extraction conditions for maximizing yield and furfurals content in peanut shells were determined as follows: temperature of 200.0 °C, extraction time of 45.0 min, and solvent-to-material ratio of 21.0 mL/g. Under these conditions, the obtained extracts showed a rich profile of biologically active compounds, including phenolic compounds and antioxidants.
