Title:Optimizacija postopka ekstrakcije sladkega pelina ( Artemisia annua L.) za izolacijo antioksidativnih in protirakavih učinkovin : diplomsko delo univerzitetnega študijskega programa I. stopnje

Abstract:Pojavnost raka se zaradi vse večje izpostavljenosti škodljivim snovem in onesnaženju okolja povečuje. Pomemben dejavnik, ki prispeva k napredovanju omenjene bolezni je oksidativni stres, katerega lahko zmanjšamo z antioksidanti. V preteklosti smo uporabljali sintetične antioksidante, danes pa se zaradi vse večje odpornosti na le te in njihovih negativnih stranskih učinkov uvajajo alternativni naravni viri. Med nje sodi tudi sladki pelin (Artemisia annua L.). Cilj diplomskega dela je bil optimizirati izolacijske postopke pridobivanja ekstraktov sladkega pelina, preveriti njihov antioksidativni potencial in določiti vpliv ekstraktov na metabolno aktivnost celične linije WM-266-4, ob nanosu. Z izvedbo nekonvencionalne ultrazvočne ekstrakcije in konvencionalnih ekstrakcijskih metod, hladne, tople in vroče maceracije ter ekstrakcije po Soxhletu, smo s topilom etanolom (W ≥ 96 %) pridobili ekstrakte sladkega pelina. Določili smo jim vsebnost antioksidantov na podlagi metode DPPH. Za najustreznejšo ekstrakcijsko metodo pridobivanja antioksidantov, toplo maceracijo z mešanjem, smo nato naredili optimizacijo eksperimentalnega načrta po računalniškem programu Design Expert. Kot raziskovani spremenljivki smo postavili temperaturo v območju med 23 °C in 80 °C ter hitrost mešanja med 100 rpm in 700 rpm. Ugotovili smo, da na vsebnost antioksidantov v ekstraktih vpliva temperatura, vpliv hitrosti mešanja med ekstrakcijo pa je zanemarljiv. Temperaturno območje od 55 °C do 62 °C se je za antioksidativni potencial ekstraktov izkazalo kot optimalno. Nazadnje smo preverili inhibicijo ekstraktov z najvišjo vsebnostjo antioksidantov na človeških melanomskih celicah s testom WST-8. Rezultati so pokazali, da vsi ekstrakti močno inhibirajo metabolno aktivnost rakavih celic, kar pripisujemo visokemu deležu antioksidantov v ekstraktih.

Title:Optimization of extraction procedure of artemisia annua l. forisolation of compounds with antioxidant and anticancerogen potential
Abstract:The incidence of cancer is increasing due to the growing exposure to harmful substances and environmental pollution. An important factor contributing to the progression of this disease is oxidative stress, which can be reduced by antioxidants. In the past, synthetic antioxidants were commonly used, but today, alternative natural sources are being introduced due to the increasing resistance to synthetic antioxidants and their negative side effects. A perspective source of bioactive compounds is sweet wormwood (Artemisia annua L.). The aim of this thesis was to optimize the extraction methods for obtaining sweet wormwood extracts, assess their antioxidant potential, and determine their impact on the metabolic activity of the WM-266-4 cell line upon application. By employing both unconventional ultrasonic extraction and conventional extraction methods such as cold, warm and hot maceration, as well as Soxhlet extraction, using ethanol solvent (W ≥ 96 %), extracts of sweet wormwood were obtained. The antioxidant properties of the extracts were determined using the DPPH method. For the most suitable method for recovery of antioxidants, warm maceration with stirring, an optimization procedure of the experimental design was carried out using the Design Expert computer program. Temperatures ranging from 23 °C to 80 °C and stirring speed between 100 rpm and 700 rpm were set as the investigated variables. It was found that the temperature significantly influenced the antioxidant content in the extracts, while the influence of stirring speed was negligible. The temperature range from 55 °C to 62 °C was identified as optimal for the intensification of antioxidant potential of the extracts. Finally, the inhibitory effects of extracts with the highest antioxidant content were tested on human melanoma cells using the WST-8 assay. The results showed that all extracts strongly inhibited the metabolic activity of cancer cells, which was attributed to high proportion of antioxidants in the extracts.
