Abstract:Namen magistrskega dela je izolacija in določitev biološko aktivnih komponent (kurkuminoidov) v ekstraktih kurkume, formulacija ekstrahiranih učinkovin s ciljem čim večje vsebnosti kurkumina in preverjanje protimikrobnega potenciala dobljenih ekstraktov in formulacij. Nastalim ekstraktom smo določili antioksidativno aktivnost z metodo DPPH· ter vsebnosti totalnih fenolov, proantocianidinov, karotenoidov in vitamina C. Ekstrakte smo nato formulirali v organogele in enkapsulate.

Title:Formulation of organogels with turmeric extracts with optimal dissolution profile
Abstract:The aim of this master thesis is the isolation and determination of biologically active components (curcruminoids) of turmeric extracts, the formulation of the extracted active compounds with the aim to maximise the curcumin content and to verify the antimicrobial potential of the produced extracts and formulations. Two different extraction methods were perfomed. In ultrasound extraction, ethanol was used as a solvent, while supercritical extraction was performed using supercritical carbon dioxide. The resulting extracts were analysed for their antioxidant activity using the DPPH method and determined for total phenols, proanthocyanidins, carotenoids and vitamin C. The extracts were then formulated into organogels and encapsulates. The curcumin content was determined in the extracts and organogels, and its solubility in supercritical carbon dioxide was tested using a high pressure cell at different pressures. Finally, an analysis of the antimicrobial potential of the prepared extracts and formulations was performed to determine the minimum inhibitory concentration. The results show that the maximum extraction yield is achieved with ultrasound extraction (5,77 %). We also spectrophotometrically determined that this extract, compared to the supercritical extract, has higher % inhibition (41,93 %) and higher content of total phenolics (2,06 mg GA/g materiala), proanthocyanidins (0,6 mg PAC/g materiala) and vitamin C (0,20 mg vitamina C/g materiala). The curcumin content is also higher in the ultrasound extracts. The ultrasound extract and its formulations have a lower minimum inhibitory concentration than the supercritical extract and its formulations. The lowest minimum inhibitory concentration (0,47 mg/mL) was determined for the ultrasound extract using Staphylococcus aureus and Candida albicans. Thus, we have further confirmed that the curcumin content is higher in the ultrasound extracts and that curcumin has antimicrobial activity.
