Title:Tehnologija zelene ekstrakcije biološko koristnih učinkovin iz alg : diplomsko delo visokošolskega strokovnega študijskega programa I. stopnje

Abstract:Diplomsko delo se osredotoča na raziskovanje in optimizacijo postopka zelene ekstrakcije biološko aktivnih spojin iz alg, s posebnim poudarkom na pridobivanju barvila fukoksantina, pomembnega karotenoida z velikim potencialom za uporabo v prehrambni in farmacevtski industriji. Eksperimentalni del je določen z predhodno obdelavo in optimizacijo pogojev superkritične ekstrakcije s pomočjo programa Design Expert, kjer smo preučevali vpliv različnih parametrov, kot sta tlak in temperatura na vsebnost fukoksantina v ekstraktih. Rezultati so pokazali, da superkritična ekstrakcija omogoča pridobivanje višjih vsebnosti fukoksantina v primerjavi z drugimi metodami. Ekstraktu, pridobljenemu s superkritično ekstrakcijo pri optimalnih pogojih (40 °C in 331 bar), ki je izkazoval najvišje vrednosti fukoksantina (52,67 mg fukoksantina/g ekstrakta), smo nadalje določili še vsebnost totalnih fenolov, proantocianidinov in antioksidantov. Poleg tega smo ocenili tudi njegov protimikrobni potencial, tako kot tudi ekstraktom, pridobljenim z metodo ultrazvočne ekstrakcije. Rezultati so pokazali, da je superkritična ekstrakcija učinkovitejša pri pridobivanju komponent, ki delujejo protimkrobno, v primerjavi z ultrazvočno ekstrakcijo, kar se je kazalo z nižjo MIC vrednostjo superkritičnih ekstraktov. S tem smo pridobili celosten vpogled v koristne lastnosti ekstrakta ter njegovo učinkovitost pri različnih aplikacijah.

Title:Technology of green extraction of biologically useful substances from algae
Abstract:The thesis focuses on the research and optimization of the green extraction process of biologically active compounds from algae, with a special emphasis on the extraction of fucoxanthin dye, an important carotenoid with great potential for use in the food and pharmaceutical industry. The experimental part is determined by preliminary treatment and optimization of supercritical extraction conditions using the Design Expert program, where we studied the influence of various parameters, such as pressure and temperature, on the fucoxanthin content in the extracts. The results show that supercritical extraction enables obtaining a higher fucoxanthin content compared to other methods. The content of total phenols, proanthocyanidins and antioxidants was further determined in the extract obtained by supercritical extraction under optimal conditions (40 °C at 331 bar), which showed the highest fucoxanthin values (52.67 mg fucoxanthin/g extract). In addition, we also evaluated its antimicrobial potential, as well as extracts obtained by the ultrasonic extraction method. The results showed that supercritical extraction is more effective in extracting the antimicrobial component compared to ultrasonic extraction, which was shown by the lower MIC value of supercritical extracts. With this, we gained a comprehensive insight into the beneficial properties of the extract and its effectiveness in various applications.
