Title:Parodontalna bolezen in mitohondrijska disfunkcija

Abstract:Uvod: Parodontalna bolezen (PB) kot posledica porušenega ravnotežja med bakterijami in gostiteljem vodi v vnetno-imunski odgovor gostitelja ter vpliva na splošno zdravstveno stanje posameznika. Oceno napredovalosti PB merimo s parodontalnimi parametri, iz katerih lahko izračunamo površino parodontalnega vnetja (PISA). Naša hipoteza je, da večje število globokih obzobnih žepov in večja PISA pomenijo višjo vrednost vnetnih biomarkerjev, kot so C-reaktivni protein (CRP), število levkocitov, predvsem nevtrofilcev, nižje razmerje med magnezijem in kalcijem, manjšo količino antioksidantov ter nižje število kopij mitohondrijske deoksiribonukleinske kisline (mtDNAcn). Materiali in metode: S pomočjo parodontalnih parametrov smo izračunali vrednost PISA. V serumu smo izmerili nivo vnetnih biomarkerjev in antioksidantov ter iz brisa lične sluznice izolirali mitohondrijsko (mtDNA) in jedrno DNA (nDNA) ter izračunali mtDNAcn. Rezultati: Večje število globokih obzobnih žepov in višjo vrednost PISA smo povezali z višjim CRP, večjim številom levkocitov, nevtrofilcev in eritrocitov, nižjim razmerjem magnezija in kalcija ter višjim nivojem sečne kisline v serumu, ne pa z nižjim mtNDAcn v brisu lične sluznice. Zaključki: Med številom globokih obzobnih žepov in vrednostjo PISA obstaja pozitivna korelacija. Vrednost PISA je ustrezen parameter za določanje vnetnega bremena PB, ni pa nujno ustrezen parameter za določanje mitohondrijske disfunkcije po izolaciji mtDNA iz brisa lične sluznice.

Title:Periodontal disease and mitochondrial dysfunction
Abstract:Introduction: Periodontal disease (PD) leads to an inflammatory-immune response of the host as a result of a disturbed balance between bacteria and the host and affects the general health of the individual. The assessment of PD progression is measured by periodontal parameters from which the size of the periodontal inflammatory surface area (PISA) can be calculated. Our hypothesis is that a greater number of deep periodontal pockets and a larger PISA means a higher value of inflammatory biomarkers such as C-reactive protein (CRP), leukocyte count, especially neutrophils, a lower magnesium to calcium ratio, a lower amount of antioxidants and a lower mitochondrial DNA copy number (mtDNAcn). Materials and methods: based on the periodontal parameters, we calculated the PISA value, measured the level of inflammatory biomarkers and antioxidants in the serum, isolated mtDNA and nuclear DNA (nDNA) from the buccal swab and calculated the number of mtDNAcn. Results: a higher number of deep periodontal pockets and a higher PISA value are associated with a higher CRP, a higher number of leukocytes, neutrophils and erythrocytes, a lower ratio of magnesium and calcium and a higher uric acid level in the serum, but not with a lower mtNDAcn in the buccal swab. Conclusions: since there is a positive correlation between the number of deep periodontal pockets and the PISA value, the PISA value is a suitable parameter for determining the inflammatory burden of PD, but not necessarily a suitable parameter for determining mitochondrial dysfunction after isolation of mtDNA from the buccal swab.
