Title:Vpliv procesnih parametrov na biosintezo in razpad klavulanske kisline : diplomsko delo univerzitetnega študijskega programa I. stopnje

Abstract:V diplomskem delu smo preučevali vpliv različnih procesnih parametrov na biosintezo in razpad klavulanske kisline v fermentacijski brozgi. Namen dela je bil sistematično oceniti, kako pH, temperatura ter začetne koncentracije klavulanske kisline, glicerola, fosfatov in nečistoč vplivajo na prirast v fermentacijskem mediju. Za načrtovanje eksperimentov smo uporabili metodo načrtovanja eksperimentov, vzorce fermentacijske brozge pa smo analizirali z metodo HPLC. Rezultati kažejo, da ima začetni pH fermentacijske brozge izrazit vpliv na biosintezo in razgradnjo klavulanske kisline, saj se z višjim začetnim pH poveča produkcija klavulanske kisline. Z naraščajočo temperaturo in koncentracijo rastnega medija se znižuje koncentracija sintetizirane klavulanske kisline. Z zviševanjem vsebnosti glicerola in fosfatov v rastnem mediju se prav tako znižuje produktivnost bakterij. Vsebnost nečistote A v mediju nakazuje na razgradnjo klavulanske kisline, zato kaže negativno korelacijo glede na prirast. Drugače je z nečistočo G, ki se je pokazala za pozitivno, saj prirast pozitivno korelira z višjo začetno koncentracijo.

Title:The influence of process parameters on the biosynthesis and degradation of clavulanic acid
Abstract:In this study, we examined the influence of various process parameters on the biosynthesis and degradation of clavulanic acid in the fermentation broth. The aim of the work was to systematically evaluate how pH, temperature, and initial concentrations of clavulanic acid, glycerol, phosphates, and impurities affect the biomass growth in the fermentation medium. We used the design of experiments (DoE) method to plan the experiments, and analyzed samples of the fermentation broth using the HPLC method. The results show that the initial pH of the fermentation broth has a pronounced effect on the biosynthesis and degradation of clavulanic acid, as a higher initial pH increases the production of clavulanic acid. Increasing temperature and growth medium concentration reduce the amount of synthesized clavulanic acid. Likewise, increasing the content of glycerol and phosphates in the growth medium reduces bacterial productivity. The presence of impurity A in the medium indicates the degradation of clavulanic acid and thus exhibits a negative correlation with biomass growth. It is different with impurity G, which proved to be beneficial, as biomass growth positively correlates with its higher initial concentration.
