Title:Vpliv različnih koncentracij kadmija na ozkolistni koščec Berula erecta Huds. v in vitro pogojih : magistrsko delo

Abstract:Vpliv kadmija (Cd) na rastline predstavlja kompleksen in pomemben vidik okoljskega onesnaženja, ki lahko resno ogroža rastline in druge organizme, ekosisteme ter pogosto presega mejne imisijske in opozorilne vrednost nekaterih območij Slovenije. Ozkolistni koščec (Berula erecta) je hitro rastoča rastlina, makrofit, ki bi se lahko uporabljala za razstrupljanje onesnaženih tal in odpadnih voda. Raziskali smo vpliv kadmija na rast in razvoj vrste Berula erecta v tkivni kulturi. Rastline so bile gojene v Murashige in Skoog gojišču z dodanimi raztopinami CdCl₂ v koncentracijah Cd; 0,1 mg L⁻¹ , 1 mg L⁻¹, 10 mg L⁻¹, 50 mg L⁻¹ in 100 mg L⁻¹. Spremljali smo rastne parametre, fotokemično učinkovitost fotosistema II, vsebnosti fotosintetskih pigmentov, antocianov, ter vsebnosti glutationa in cisteina v rastlinskih tkivih. Dodatek kadmija v koncentracijah med 0,1 mg L⁻¹ in 10 mg L⁻¹ ni vplivala na rast in razvoj vrste Berula erecta. Zaviralni učinek na rast rastlin, fotosintetske pigmente klorofile, karotenoide in antociane se je izrazil pri koncentracijah 50 mg L⁻¹ Cd in 100 mg L⁻¹ Cd, kar se je odražalo v zmanjšanju višine in rasti korenin in zmanjšano rast stolonov in brstov. Koncentraciji glutationa (GSH) in cisteina (CYS) sta se z naraščajočo koncentracijo Cd zmanjšali, kar kaže na moteno redoks ravnovesje v celici. Ugotavljamo, da je Berula erecta primeren modelni organizem za biomonitoring Cd v vodnih ekosistemih, saj njeni rastni in biokemijski parametri hitro in občutljivo reagirajo na prisotnost te težke kovine.

Title:Effect of various cadmium concentrations on Berula erecta Huds. in in vitro conditions
Abstract:The effect of cadmium (Cd) on plants is a complex and important aspect of environmental pollution that can be a serious threat to plants, other organisms and ecosystems and frequently exceeds the immission and warning limits in some areas of Slovenia. Lesser water-parsnip (Berula erecta) is a fast growing macrophyte plant, which can be used for detoxification of polluted soil and wastewater. The effect of cadmium on the growth and development of Berula erecta in tissue culture was studied. Plants were grown in Murashige and Skoog medium enriched with cadmium in the form of CdCl₂ at concentrations 0,1 mg L-¹, 1 mg L-¹, 10 mg L-¹, 50 mg L-¹ and 100 mg L-¹. We analysis the changes in growth parameters, the photochemical efficiency of photosystem II, the content of photosynthetic pigments, anthocyanins, the glutathione and cysteine content in plant tissues. Cadmium had no effect on the growth and development of Berula erecta at concentrations between 0,1 mg L⁻¹ and 10 mg L⁻¹. The inhibitory effect on plant growth exhibited at concentrations cadmium of 50 mg L-1 and 100 mg L-1, resulting in a reduction in root height and growth, while at the same time stimulating the growth of stolons and buds.
