Title:Določanje vsebnosti rastlinskim hormonom podobnih substanc v žlezavi nedotiki (lmpatiens glandulifera) s pomočjo biotestov : magistrsko delo

Abstract:Žlezava nedotika ($\itImpatiens glandulifera$) je invazivna tujerodna vrsta, ki se hitro razširja po več tipih habitatov. Hitrost njenega razširjanja in rasti bi lahko bila povezana s hitro rastjo in rastlinskim hormonom podobnih substanc. Cilj naše naloge je bil z biotesti raziskati vsebnost rastlinskim hormonom podobnih substanc ločeno v listih in koreninah, in sicer v šestih razvojnih stadijih življenjskega cikla. Uporabili smo tri bioteste: $\itTriticum$, $\itMung$ in $\itCucumis$ biotest. S $\itTriticum$ biotestom smo določali vsebnost citokininom podobnih substanc. Z $\itMung$ biotestom smo določali vsebnost avksinom podobnih substanc. S $\itCucumis$ biotestom smo določali vsebnost obeh skupin rastlinskim hormonom podobnih substanc, tako citokininskih in avksinskih. Citokininski učinki v žlezavi nedotiki, določeni s $\itTriticum$ biotestom, so bili primerljivi z učinkom vsebnosti 0,5 mg L⁻¹ do 4 mg L⁻¹ 6-benzilamonopurina (BAP), s $\itCucumis$ biotestom pa učinku vsebnosti do 0,5 mg L⁻¹ BAP. Najvišje primerljive vsebnosti citokininom podobnih substanc smo zaznali v drugem razvojnem stadiju življenjskega cikla, ko so rastline intenzivno rasle v višino, cvetovi pa še niso bili razviti ter v zadnjem razvojnem stadiju s popolnoma razvitimi plodovi in semeni. Avksinski učinek, določen s $\itCucumis$ biotestom, je bil primerljiv z učinkom vsebnosti 0,5 mg L⁻¹ do 2 mg L⁻¹ indol-3-maslene kisline (IBA) in z $\itMung$ testom primerljiv z učinkom vsebnosti 0 mg L⁻¹ do 8 mg L⁻¹ IBA. Najvišje vsebnosti avksinom podobnih substanc so bile prisotne v prvem razvojnem stadiju, torej v kalicah, ter v zadnjem razvojnem stadiju življenjskega cikla, ko je rastlina razvila razmnoževalne strukture.

Title:Determination of plant hormone-like substances of the himalayan balsam (Impatiens glandulifera) with bioassays
Abstract:Himalayan balsam ($\itImpatiens glandulifera$) is an invasive alien species known for its rapid growth and efficient spread across diverse habitats. Its invasive success may be linked to high growth rates and the presence of plant hormone-like substances. This study aimed to investigate, using bioassays, the occurrence and distribution of plant hormone-like substances in leaves and roots across six developmental stages of the life cycle. Three bioassays were used: the $\itTriticum$ bioassay to assess cytokinin-like activity, the $\itMung$ bean bioassay to assess auxin-like activity, and the $\itCucumis$ bioassay to assess both cytokinin- and auxin-like activities. Cytokinin-like effects detected in $\itI. glandulifera$ using the $\itTriticum$ bioassay were comparable to those induced by 0,5–4 mg L⁻¹ 6-benzylaminopurine (BAP), while the $\itCucumis$ bioassay indicated effects comparable to concentrations of up to 0,5 mg L⁻¹ BAP. The highest levels of cytokinin-like activity were observed during the second developmental stage, characterized by intensive vegetative growth before flowering, and during the final developmental stage, when fruits and seeds were fully developed. Auxin-like activity determined using the $\itCucumis$ bioassay was comparable to the effects of 0,5–2 mg L⁻¹ indole-3-butyric acid (IBA), while the $\itMung$ bean bioassay indicated activity comparable to 0–8 mg L⁻¹ IBA. The highest levels of auxin-like substances were detected during the first developmental stage (seedlings) and during the final developmental stage, corresponding to the formation of reproductive structures.
