Title:Iskanje novih molekularnih označevalcev pri polipih debelega črevesa in danke s sodobnimi pristopi bioinformatike

Abstract:CRC je eden izmed najpogostejših in najbolj smrtonosnih rakov. Kljub napredkom v diagnosticiranju in zdravljenju ostajajo izzivi zaradi njegove heterogenosti. Zgodnje odkrivanje raka z uporabo molekularnih označevalcev bi lahko bistveno izboljšalo njegovo zdravljenje in znižalo smrtnost. V okviru presejalnega programa SVIT smo zbrali številne vzorce polipov in zdrave sluznice. S pomočjo bioinformatskega programa Cytoscape smo pridobili seznam kandidatnih genov in RNA. Nato je bila izvedena izolacija RNA, priprava cDNA in analiza genske ekspresije kandidatnih genov ter RNA z metodo RT-PCR. Na koncu smo s pomočjo SPSS, programa za statistično obdelavo podatkov, pridobljene podatke še analizirali. Analiza podatkov je pokazala statistično značilne razlike v izražanju genov NETO2, CDK6, AMMECR1, ADH1B, CDKN2B, MYH11, AKR1B10, SMPDL3A in RNA miR-1-3p ter miR-124-3p. Izmed teh imajo NETO2, CDK6 in AMMECR1 potencial za terapevtsko tarčo. Preostali geni in RNA niso imeli statistično značilnih razlik, ali analiza zaradi neizražanja ni uspela. Rezultati podpirajo dejstvo, da je uporaba bioinformatskih metod pomembna v zgodnjem odkrivanju CRC in njegovem zdravljenju. Predstavlja potencial za odkrivanje novih načinov diagnosticiranja in zdravljenja te bolezni.

Title:Identification of new molecular markers in the polyps of colon and rectum, using modern bioinformatics approaches
Abstract:CRC is one of the most common and deadliest cancers. Despite all the advancements in diagnostics and treatment, there are still many challenges due to its heterogeneity. Its early detection, using molecular biomarkers, could significally improve treatment outcome and reduce mortality. Numerous polyps and healthy tissue samples were collected with the help of SVIT screening program. Using Cytoscape, we created a list of candidate genes and RNA. We isolated RNA from our samples, prepared cDNA and, with the use of RT-PCR, analyzed gene expression. Finally, we analyzed the data using SPSS. Data analysis revealed statistically significant differences in the expression of genes NETO2, CDK6, AMMECR1, ADH1B, CDKN2B, MYH11, AKR1B10, SMPDL3A, as well as RNA miR-1-3p and miR-124-3p. NETO2, CDK6 and AMMECR1 also have potential as therapeutic targets. The remaining genes and RNA showed no statistically significant differences, or could not be analyzed due to low expression. The results support the importance of using bioinformatics methods in the early detection of CRC and its treatment. It has the potential to develop new diagnostic and therapeutic strategies for the disease.
