Title:Genetska identifikacija zgodovinskih sort vinske trte (v. vinifera subsp. vinifera) na štajerskem in divje trte (v. vinifera subsp. sylvestris) v sloveniji : doktorska disertacija

Abstract:Ohranjanje in karakterizacija genskih virov vinske trte sta ključnega pomena za razumevanje vinogradniške zgodovine ter za podporo trajnostni pridelavi grozdja. V Sloveniji so bile tako zgodovinske sorte vinske trte kot tudi populacije divje vinske trte Vitis vinifera L. subsp. sylvestris dlje časa obravnavane kot nezadostno dokumentirane ali celo odsotne. Namen te doktorske raziskave je bil identificirati in genetsko karakterizirati zgodovinske sorte vinske trte, ohranjene v starih vinogradih, ter potrditi prisotnost, analizirati gensko strukturo in stanje ohranjenosti populacij divje vinske trte v Sloveniji. Med letoma 2019 in 2024 so bile izvedene obsežne terenske raziskave v starih vinogradih in naravnih rastiščih. Skupno je bilo zbranih 340 akcesij vinske trte s 115 lokacij starih vinogradov v slovenskem vinorodnem okolišu Štajerska, ki so bile analizirane z uporabo 24 SSR-markerjev. Genske analize so razkrile 68 različnih genotipov, med njimi 29 potrjenih zgodovinskih sort in 39 predhodno neznanih avtohtonih genotipov, kar potrjuje obstoj pred filokserne genske raznolikosti vinske trte in situ. Analize genske sorodnosti so pokazale tesne genealoške povezave med več zgodovinskimi sortami, kar poudarja njihov zgodovinski pomen v regionalnem sortimentu vinske trte. Vzporedno so bile v Sloveniji prvič sistematično popisane in genetsko karakterizirane populacije divje trte. Skupno je bilo na petih naravnih rastiščih evidentiranih 126 akcesij divje trte, od tega je bilo 89 akcesij analiziranih z uporabo SSR-markerjev in markerjev za določanje spola cvetov. Rezultati so pokazali, da slovenska populacija sylvestris predstavlja genetsko jasno ločeno in večinoma čisto populacijo, izrazito diferencirano od gojenih sort V. vinifera in podlag, z omejenimi dokazi o introgresiji. Populacijske analize so pokazale, da je slovenska populacija divje vinske trte genetsko najsorodnejša balkanskim in srednjeevropskim skupinam sylvestris. Raziskava predstavlja prve celovite genetske dokaze o ohranjenosti zgodovinske sortne pestrosti vinske trte in avtohtonih populacij divje vinske trte v Sloveniji. Rezultati poudarjajo nujnost sistematičnega izvajanja ukrepov ohranjanja in situ in ex situ ter izpostavljajo pomen teh genskih virov za prihodnje žlahtnjenje in trajnostni razvoj vinogradništva.

Abstract:Conservation and characterization of grapevine genetic resources are crucial for understanding viticultural history and supporting sustainable grape production. In Slovenia, both historical grapevine varieties and populations of the wild grapevine (Vitis vinifera L. subsp. sylvestris) have long been considered insufficiently documented or even absent. The aim of this doctoral research is to identify and genetically characterize historical grapevine varieties preserved in old vineyards and to confirm the presence, genetic structure, and conservation status of wild grapevine populations in Slovenia. Between 2019 and 2024, extensive field research was conducted in old vineyards and natural habitats. A total of 340 grapevine accessions were collected from 115 old-vineyard locations in the Slovenian Styria wine-growing district, and these were analyzed using 24 SSR markers. Genetic analyses revealed 68 distinct genotypes, including 29 confirmed historical varieties and 39 previously unknown indigenous genotypes, confirming the existence of pre-phylloxera genetic diversity in situ. Genetic relationship analyses showed close genealogical links between several historical varieties, highlighting their historical significance in the regional assortment. In parallel, wild grapevine populations in Slovenia were systematically surveyed and genetically characterized for the first time. A total of 126 wild grapevine accessions were recorded across five natural habitats, of which 89 were analyzed using SSR markers and flower sex-determination markers. Results showed that the Slovenian sylvestris population represents a genetically distinct and largely pure population, clearly differentiated from cultivated V. vinifera varieties and rootstocks, with limited evidence of introgression. Population analyses indicated that the Slovenian wild grapevine population is genetically most closely related to Balkan and Central European sylvestris groups. This research provides the first comprehensive genetic evidence for the preservation of historical grapevine variety diversity and indigenous wild grapevine populations in Slovenia. The results emphasize the necessity of implementing systematic in situ and ex situ conservation measures and highlight the importance of these genetic resources for future breeding and the sustainable development of viticulture.
