Title:Optimizacija ekstrakcije bioaktivnih spojin iz lanene pogače z Randallovo metodo

Abstract:V diplomskem delu smo preučevali ekstrakcijo bioaktivnih spojin iz lanene pogače z Randallovo metodo. Namen dela je bil ovrednotiti vpliv procesnih parametrov kot so čas ekstrakcije, temperatura, razmerje med topilom in surovino ter sestava ekstrakcijskega topila na lastnosti pridobljenih ekstraktov. Ekstrakcije smo izvedli z mešanico etanola in vode v skladu s predhodno zastavljenim eksperimentalnim načrtom. Pridobljene ekstrakte smo ovrednotili na podlagi masnega izkoristka ekstrakcije, vsebnosti skupnih fenolnih spojin, flavonoidov, skupnega organskega ogljika in antioksidativne aktivnosti. Rezultate smo statistično obdelali in modelirali v programu Design Expert z uporabo metode odzivnih površin na osnovi Box Behnkenovega načrta poskusov. Rezultati so pokazali, da procesni parametri signifikantno vplivajo na ekstrakcijo bioaktivnih spojin iz lanene pogače, pri čemer višji masni izkoristek ni nujno pomenil višje vsebnosti bioaktivnih spojin. Vpliv posameznih parametrov je bil odvisen tudi od medsebojnih interakcij. Sestava topila (delež etanola v topilu) je pomembno vplivala na lastnosti ekstraktov, zato je izbira ekstrakcijskega topila ključna pri usmerjeni ekstrakciji posameznih spojin. Na podlagi analize eksperimentalnih podatkov so bili kot najustreznejši pogoji za ekstrakcijo bioaktivnih spojin iz vzorcev lanene pogače z Randallovo metodo določeni čas ekstrakcije 1,07 h, temperatura 81,73 °C in razmerje topilo : surovina 18,20 mL/g surovine. Pri teh pogojih je model napovedal masni izkoristek ekstrakcije 21,90 %, vsebnost skupnih fenolnih spojin 4,14 mg GAE/g suhe lanene pogače in antioksidativno aktivnost 8,38 % inhibicije DPPH radikala. Ugotovili smo, da je Randallova metoda primerna za pridobivanje ekstraktov iz lanene pogače, pri čemer je za doseganje ugodnih lastnosti ekstrakta pomembna ustrezna izbira ekstrakcijskih pogojev.

Title:Optimization of the extraction of bioactive compounds from flaxseed cake using the Randall method
Abstract:This thesis investigated the extraction of bioactive compounds from flaxseed cake using the Randall method. The aim of the study was to evaluate the effects of process parameters such as extraction time, temperature, solvent to solid ratio, and extraction solvent composition on the properties of the obtained extracts. The extractions were performed using an ethanol water mixture according to a predefined experimental design. The obtained extracts were evaluated based on mass extraction yield, total phenolic content, flavonoid content, total organic carbon, and antioxidant activity. The results were statistically analysed and modelled in Design Expert software using response surface methodology based on a Box Behnken experimental design. The results showed that the process parameters significantly affected the extraction of bioactive compounds from flaxseed cake, while a higher mass extraction yield did not necessarily correspond to a higher content of bioactive compounds. The effects of the individual parameters also depended on their interactions. The solvent composition, expressed as the ethanol fraction in the solvent, had a significant effect on the properties of the extracts; therefore, the selection of an appropriate extraction solvent is essential for the targeted extraction of specific compounds. Based on the analysis of the experimental data, the most suitable conditions for the extraction of bioactive compounds from flaxseed cake using Randall method were determined to be an extraction time of 1,07 h, a temperature of 81,73 °C, and a solvent to solid ratio of 18,20 mL/g of raw material. Under these conditions, the model predicted a mass extraction yield of 21,90 %, a total phenolic content of 4,14 mg GAE/g of dry flaxseed cake, and an antioxidant activity of 8,38 % DPPH radical inhibition. The results demonstrated that the Randall method is suitable for obtaining extracts from flaxseed cake and that the appropriate selection of extraction conditions is essential for achieving favourable extract properties.
