Title:Vpliv ekstrakcijskih metod na vsebnost bioaktivnih spojin iz navadne kislice (Rumex acetosa)

Abstract:Namen diplomskega dela je bil raziskati, kako različne ekstrakcijske metode, topila in pogoji vplivajo na vsebnost bioaktivnih spojin v vzorcih, pridobljenih iz navadne kislice (Rumex acetosa). Preizkusili smo hladno in vročo maceracijo, ultrazvočno ekstrakcijo ter ekstrakcijo s superkritičnim CO2. Pri maceraciji in ultrazvočni ekstrakciji smo kot topila uporabili etanol, metanol, aceton in vodo. Superkritično ekstrakcijo smo izvedli pri dveh temperaturah (40 °C in 60 °C). Ekstraktom smo spektrofotometrično določili antioksidativno aktivnost ter vsebnost totalnih fenolov in proantocianidinov. Izkoristki maceracije in ultrazvočne ekstrakcije so bili nizki, med 4,67 % in 10,26 %, pri čemer so bili najvišji pri uporabi 70 % metanola. Izkoristki vroče maceracije so bili višji od izkoristkov hladne maceracije, prednosti ultrazvoka pa so se pokazale predvsem pri vodni ekstrakciji. Antioksidativno aktivnost smo podali kot odstotek radikalske inhibicije, pri čemer so bile večje razlike opazne pri uporabi različnih topil kot pri različnih ekstrakcijskih metodah. Pri vseh mešanicah organskih topil z vodo so bile vrednosti inhibicije nad 80 %. Vsebnost totalnih fenolov in proantocianidinov smo izrazili na gram ekstrakta ter gram materiala. V vseh primerih so bile najvišje koncentracije pri vzorcih vroče maceracije s 70 % acetonom. Preostali trendi rezultatov so se razlikovali glede na ekstrakcijsko metodo in uporabljeno topilo. Superkritična ekstrakcija ni pokazala prednosti, ki jih po navadi pripisujemo tej metodi. Izkoristka sta bila med najnižjimi, nekoliko višji je bil pri 60 °C. Rezultate analiz težko primerjamo z ostalimi vzorci zaradi drugačnega agregatnega stanja ekstraktov. Poleg tega je pridobivanje zadostne količine ekstraktov zahtevalo daljše čase ekstrakcije, za ekstrakcijo bolj polarnih spojin pa je bila potrebna uporaba kotopila (etanola).

Title:Effect of extraction methods on the content of bioactive compounds from common sorrel (Rumex acetosa)
Abstract:The purpose of this thesis was to investigate how different extraction methods, solvents, and conditions affect the content of bioactive compounds in samples obtained from common sorrel (Rumex acetosa). Cold and hot maceration, ultrasound‑assisted extraction, and extraction with supercritical CO2 were evaluated. Ethanol, methanol, acetone, and water were used as solvents for maceration and ultrasound‑assisted extraction. Supercritical extraction was performed at two temperatures (40 °C and 60 °C). The antioxidant activity, total phenolic content, and proanthocyanidin content of the extracts were determined using spectrophotometry. The yields of maceration and ultrasound‑assisted extraction were low, ranging from 4.67 % to 10.26 %, with the highest yields obtained using 70 % methanol. Hot maceration resulted in higher extraction yields than cold maceration, while the advantages of ultrasound were most evident in aqueous extraction. Antioxidant activity was expressed as the percentage of radical inhibition, where greater differences were observed depending on the solvent used rather than the extraction method. For all mixtures of organic solvents with water, inhibition values exceeded 80 %. The content of total phenols and proanthocyanidins was expressed per gram of extract and per gram of plant material. In all cases, the highest concentrations were found in samples obtained using hot maceration with 70 % acetone. Other result trends varied according to the extraction method and solvent used. Supercritical extraction did not demonstrate the typical advantages usually attributed to this method, with yields among the lowest and only slightly higher at 60 °C. The analysis results are difficult to compare with other samples due to the different physical state of the extracts. Furthermore, obtaining a sufficient quantity of extracts required longer extraction times, and the use of a co-solvent (ethanol) was necessary for the extraction of more polar compounds.
