Title:Biokemijska označitev izolatov pseudomonas spp. iz lezij listov murve (morus sp.)

Abstract:Bakterije rodu Pseudomonas vključujejo številne fitopatogene vrste, ki povzročajo gospodarsko pomembne bolezni rastlin. V diplomskem delu je predstavljena biokemijska označitev bakterijskih izolatov rodu Pseudomonas, pridobljenih iz lezij listov murve (Morus sp.). Eksperimentalni del je obsegal klasične biokemijske teste za ugotavljanje aktivnosti encimov želatinaze in arginin dihidrolaze ter sposobnosti razgradnje ogljikovih hidratov (glukoze, ksiloze, adonitola in manitola) ter tvorbo levana. Sintezo levana smo dodatno potrdili z analizo produktov kislinske hidrolize s tankoplastno kromatografijo (TLC). Rezultati so pokazali, da noben izmed testiranih izolatov Pseudomonas spp. ne razgrajuje želatine in ne sintetizira encima arginin dihidrolaze. Vsi izolati so bili sposobni presnavljati glukozo v aerobnih in anaerobnih razmerah, medtem ko adonitola ni razgrajeval nobeden izmed testiranih izolatov. Pri razgradnji manitola in ksiloze smo opazili raznolikost v biokemijskih lastnostih posameznih izolatov. Slovenski izolat Pseudomonas amygdali pv. mori, izolat MB01, teh dveh sladkorjev ni bil sposoben presnavljati, medtem ko so ga preostali trije izolati (Pseudomonas caspiana izolat KP01, Pseudomonas caspiana izolat KK01, Pseudomonas amygdali pv. mori izolat Ma001) lahko. S kromatografsko analizo TLC smo pri vseh izolatih uspešno potrdili sintezo zunajceličnega polisaharida levana, saj so se lise hidroliziranih vzorcev ujemale z referenčno liso fruktoze. Rezultati nakazujejo možno prilagajanje lokalnih populacij bakterij specifičnim mikrookoljskim razmeram ter potrjujejo, da biokemijska označitev sama po sebi ni zadostna za zanesljivo razlikovanje fitopatogenih psevdomonad, zato jo je smiselno dopolniti z analizo genetskih označevalcev.

Title:Biochemical characterization of pseudomonas spp. isolates from lesions of mulberry (morus sp.) leaves
Abstract:Bacteria belonging to the genus Pseudomonas include numerous phytopathogenic species that cause economically important plant diseases. This thesis presents the biochemical characterization of Pseudomonas spp. isolates obtained from lesions on mulberry (Morus sp.) leaves. The experimental work included classical biochemical tests to determine gelatinase and arginine dihydrolase activity, the ability to utilize carbohydrates (glucose, xylose, adonitol, and mannitol), and levan synthesis. Levan synthesis was further confirmed by analyzing the products of acid hydrolysis using thin-layer chromatography (TLC). The results showed that none of the tested isolates hydrolyzed gelatin or exhibited arginine dihydrolase activity. All isolates were capable of utilizing glucose under both aerobic and anaerobic conditions, whereas none of them utilized adonitol. Diversity in the biochemical characteristics of the isolates was observed in the utilization of mannitol and xylose. The Slovenian isolate Pseudomonas amygdali pv. mori MB01 was unable to utilize either of these carbohydrates, while the remaining three isolates (Pseudomonas caspiana KP01, Pseudomonas caspiana KK01, and Pseudomonas amygdali pv. mori Ma001) were able. TLC analysis confirmed the synthesis of the extracellular polysaccharide levan in all isolates, as the spots obtained from hydrolyzed samples corresponded to the reference fructose spot. The results suggest possible adaptation of local bacterial populations to specific microenvironmental conditions and confirm that biochemical characterization alone is insufficient for reliable differentiation of phytopathogenic pseudomonads. Therefore, it should be complemented by the analysis of genetic markers.
