Title:Genetska analiza kandidatnega področja na kromosomu 10 pri bolnikih s kronično vnetno črevesno boleznijo

Abstract:Glavna podtipa kronično vnetne črevesne bolezni (KVČB) sta Crohnova bolezen (CB), za katero so značilna kronična vnetja v celotni prebavni cevi vključno s tankim črevesom in ulcerozni kolitis (UC), kjer je vnetje omejeno pretežno na široko črevo in danko. Primerjalne študije so pokazale, da k razvoju KVČB poleg dejavnikov okolja veliko prispeva tudi genetski faktor. Namen magistrskega dela je bil ugotoviti povezave polimorfizmov posameznega nukleotida (SNP) na kromosomu 10 s tveganjem za nastanek bolezni pri slovenskih bolnikih s KVČB. Pri raziskavi smo uporabili različne bioinformatske pristope za izbiro potencialnih kandidatnih SNP-jev in genov vključno z analizami asociacijskih študij na celotnem genomu (GWA) ter različne tehnike iz molekularne biologije za laboratorijske analize. Potencialne kandidatne SNP-je smo izbrali iz 16-ih GWA študij, v katerih je bilo potrjenih 13 SNP-jev na kromosomu 10 povezanih s KVČB. Za asociacijsko analizo smo izbrali tri SNP-je iz področja 10p in dva SNP-ja iz področja 10q. Asociacijska analiza, pri kateri smo genotipizacijo izbranih SNP-jev izvedli na 162 zdravih posameznikih in 558 bolnikih s KVČB, je pokazala povezavo med KVČB in SNP-ji rs4934697, rs10509115, rs12777960 in rs2254252. V nadaljnji analizi smo izvedli ekspresijsko študijo izoforme 1 gena CCNY in izoforme 1 gena CREM, ki sta sosednja gena SNP-jev rs12777960 in rs4934697, vendar povezave med geni in izbranima SNP-oma nismo ugotovili. Vsekakor na področju genetike KVČB niso pojasnjeni vsi dejavniki, ki vplivajo na tveganje za razvoj KVČB, vendar pa lahko z odkrivanjem in določanjem novih SNP-jev ter genov, ki povečujejo dovzetnost za razvoj KVČB, omogočimo lažjo napoved bolezni kot tudi lažje načrtovanje diagnostično terapevtskih postopkov in posegov.

Title:Genetic analysis of the chromosome 10 candidate region in patients with inflammatory bowel disease
Abstract:The main subtypes of inflammatory bowel disease (IBD) are Crohn's disease (CD), characterized by chronic inflammations of the entire gastrointestinal tract together with small intestine and ulcerative colitis (UC), where inflammation is mostly limited to the colon and rectum. Comparative studies have shown that the genetic factor contributes a great part of the cause for IBD. The aim of this master's dissertation is to discover single nucleotide polymorphisms (SNP) on the chromosome 10 with susceptibility to IBD in Slovene patients with IBD. We used different bioinformatics approaches for choosing a potential candidate SNPs and genes together with genome wide association studies (GWA) and different techniques from molecular biology for analysis of selected SNPs and genes. We have chosen potential SNP candidates out of 16 GWA studies in which 13 SNPs were confirmed on the chromosome 10 to be associated with IBD. For association analysis we chose 3 SNPs from the locus 10p and 2 SNPs from locus 10q. In association analysis we genotyped selected SNPs in 162 healthy individuals and 558 patients with IBD. We found association between IBD and SNPs rs4934697, rs10509115, rs12777960, and rs2254252. Further we performed an expression study of isoform 1 of gene CCNY and isoform 1 of gene CREM. Those two genes are located next to SNPs rs12777960 and rs4934697. We didn’t find any association between genes and chosen SNPs. In the field of genetics of IBD, all factors that effect the risk for IBD development are not explained, but by discovering and determining new SNPs and genes, that increase susceptibility to develop IBD, we can enable an easy projection of a disease as well as easier planning of a diagnostic-therapeutic procedures and interventions.
