Title:ISOLATION AND IDENTIFICATION OF BACTERIOCINOGENIC STRAIN OF LACTOBACILLUS PLANTARUM WITH POTENTIAL BENEFICIAL PROPERTIES FROM DONKEY MILK

Abstract:Abstract: The goal of this study was to isolate and characterize LAB (lactic acid bacteria) with potential beneficial properties for human health. LAB were isolated from donkey milk and characterized based on physiological, biochemical and molecular methods. The isolate that presented highest bacteriocin potential (Lact. plantarum LP08AD) was evaluated for the production of bacteriocin. Bactericidal effect of bacteriocin produced by LP08AD on Listeria. monocytogenes, Enterococus. faecium and Lact. curvatus was shown for actively growing and stationary cells. Bacteriocin from LP08AD adhered at low levels to the producer cells. Presence of plantaricin W gene on the genomic DNA was recorded based on PCR. Good growth of strain LP08AD was recorded in the pH range from 5.0 to 9.0 and LP08AD grew well in the presence of bile acids in concentration below 0.8%. LP08AD demonstrated low attachment ability to the small intestinal epithelial polarised monolayers of H4, PSI and CLAB, with values similar for cells of human and pig origin. In addition, LP08AD showed ability to remove cholesterol. Bacteriocin-producing LP08AD might be useful in the design of novel functional foods with potential probiotic or biopreservation properties.

Title:IZOLACIJA IN IDENTIFIKACIJA BAKTERIOCIN PROIZVAJAJOČIH SEVOV Lactobacillus Plantarum IZ OSLIČJEGA MLEKA
Abstract:Izvleček: Namen šudije je bil izolirati in okarakterizirati mlečnokislinske bakterije (LAB), ki bi lahko imele potencialne koristne lastnosti za zdravje človeka. Identifikacija LAB iz oslovskega mleka je potekala s pomočjo fizioloških, biokemičnih in molekularnih metod. Ovrednotena je bila proizvodnja bakteriocinov tistega izoliranega seva LAB, ki je pokazal najvišjo bakteriocinsko aktivnost (Lactobacillus plantarum LP08AD). Pri testiranju se je bakteriocidna aktivnosti bakteriocina iz LP08AD pojavila tako pri rastočih kot tudi pri stacionarnih celičnih kulturah testnih sevov Listeria. monocytogenes, Enterococus. faecium in Lact. curvatus. Majhne količine bakteriocina LP08AD so bile prisotne tudi na lastnih proizvodnih celicah. Z uporabo metode PCR se je na nivoju genomske DNA dokazala prisotnost gena za plantaricin W. Dobra rast seva LP08AD je bila zaznana pri pH od 5,0 do 9,0, prav tako ob prisotnosti žolčnih kislin v koncentraciji manjši od 0,8%. Bakterije LP08AD so bile nanešene tudi na enoslojne polarizirane epitelijske celične kulture H4, PSl in CLAB. Vrednosti pritrjenih celic seva LP08AD so bile nizke pri vseh preučevanih celičnih linijah in s podobnimi izidi, ne glede na humani ali prašičji izvor celičnih linij. LP08AD sev je pokazal zmožnost zniževanja ravni holesterola. Bakteriocin izražajoči sev LP08AD ima tako velik potencial pri razvoju novih prehranskih dopolnil s probiotičnimi in bio-konzervacijskimi lastnostmi.
