Title:Razgradnja skatola s pomočjo mikroorganizmov kot alternativa zmanjševanja vonja po merjascu

Abstract:Čeprav ima reja merjascev veliko prednosti v primerjavi s kastrati, je v številnih državah kastracija mladih prašičev ustaljena praksa, predvsem zaradi preprečevanja razvoja vonja po merjascu. Eden glavnih vzrokov za vonj po merjascu je akumulacija androstenona in skatola v maščobnem tkivu. Naravni način odstranjevanja neprijetnega vonja z uporabo mikroorganizmov bi lahko bil alternativa obstoječim metodam. Namen magistrske naloge je bil iz blata merjasca izolirati mikroorganizme, ki so odporni na toksičnost skatola in so ga sposobni tudi razgraditi. S pomočjo metode difuzije na agarnih ploščah je bilo izoliranih 20 kolonij mikroorganizmov, ki so rasli v coni inhibicije skatola. Predvideli smo, da so izolirani mikroorganizmi odporni na visoke koncentracije skatola, za kar bi lahko bil vzrok tudi njegova aktivna razgradnja. V nadaljevanju smo s pomočjo spektrofotometričnega določanja skatola ugotavljali, ali so izolirani mikroorganizmi sposobni razgraditi skatol. Zaradi težav pri izvedbi metode nismo uspeli nedvoumno potrditi, da je katerikoli od izoliranih sevov sposoben metabolizma skatola.

Title:Degradation of skatole by microorganisms as an alternative to diminish boar taint
Abstract:Although rearing of entire males has a number of advantages compared to castrated male pigs, in most countries the castration of young pigs is an established practice, mainly to avoid the potential problem of boar taint. The main cause for boar taint is accumulation of androstenone and skatole in fat tissue. Removing the unpleasant odour by using microorganisms could be a natural way and an alternative to existing methods. The purpose of the master thesis was to isolate microorganisms, resistant to the toxicity of skatole and with a potential to degrade it. Using agar well diffusion method, 20 colonies of microorganisms, which grew in the inhibition zone of skatole, were isolated. We assumed, that isolated microorganisms are resistant to high concentrations of skatole, where a potential mechanism of resistance could also be its active degradation. With the spectrophotometric method for determination of skatole, we examined the isolated microorganisms on their capability of skatole degradation. Due to the difficulties in method implementation we could not unequivocally confirm that any of the isolated strains is capable of skatole metabolism.
