Title:Študija proizvodnosti hidrolize saharoze z encimom vezanim na odpadna steklena vlakna : magistrsko delo

Abstract:V raziskavi smo kot nosilec za imobilizacijo encima invertaze uporabili odpadna steklena vlakna. Encim smo kovalentno vezali na modificirani nosilec. Učinkovitost imobilizacije smo testirali na reakciji hidrolize saharoze. Vse eksperimente smo izvajali v laboratorijskem šaržnem reaktorju. Preučevali smo vpliv pH (4,5, 5, 5,5 in 6) in reakcijske temperature (20, 25, 30 in 35) °C na presnovo reakcije. Učinkovitost reakcije smo spremljali in-situ z merjenjem koncentracije saharoze, glukoze in fruktoze z uporabo FTIR sonde. Na koncu smo preverili vpliv pH (4,5, 5, 5,5 in 6) imobilizacije na presnovo pri predhodno določenih optimalnih pogojih reakcije. Dosegli smo visoko učinkovitost imobilizacije. Določili smo optimalen pH reakcijske mešanice in temperaturo reakcije. Nadalje smo ob večkratni zaporedni uporabi katalizatorja določili aktivnost invertaze, imobilizirane na steklena vlakna. Raziskavo smo nadgradili s kinetično študijo reakcije. Iz enačbe encimske kinetike smo določili konstante proizvodnosti ter z uporabo Arrhenius-ove zveze aktivacijsko energijo in pred-eksponentni faktor.

Title:Study of reaction rate of sucrose hydrolysis with enzyme bounded to the waste glass fibre
Abstract:In this study we have used waste glass fibers as a carrier for immobilization of enzyme invertase. The enzyme was bound to a modified carrier by a covalent bond. .Efficiency of immobilization was tested on sucrose hydrolysis reaction. All experiments were conducted in laboratory batch reactor. Effect of pH (4,5, 5, 5,5 in 6) and reaction temperature (20, 25, 30 in 35) °C on reaction yield was explored. Reaction effectiveness was monitored in-situ by measuring sucrose, fructose and glucose concentration using FTIR probe. In the end, we have explored the effect of pH (4,5, 5, 5,5 in 6) of immobilization at determined optimal conditions on reaction yield. We have achieved high effectiveness of immobilization. We determined optimal pH value of reaction mixture and reaction temperature. Furthermore, the activity of invertase immobilized to fiberglass was determined by multiple consecutive use of enzyme. The study was advanced by kinetic study of reaction. We have determined the productivity constants with enzyme kinetic equation. We have also determined activation energy and pre-exponential factor from Arrhenius plot.
