Abstract:Perutninsko perje predstavlja enega najbolj obremenilnih in neizkoriščenih odpadkov, saj zavzema do 10 % celotne teže perjadi, kar na letni ravni nanese skoraj 10 milijonov ton odpadkov.

Title:Isolation of keratin from feathers using hydrothermal processes
Abstract:Poultry feathers are among the most polluting and unexploited wastes. Feathers represent up to 10 % of chicken weight, which leads to 10 million tons of waste annually. Because of high keratin content (90 %), feathers are a potential raw material for obtaining highly-valuable protein. Therefore, feathers could become a valuable resource, rather than polluting waste. As a part of master’s degree, isolation of keratin from chicken feathers with subcritical water at different temperatures (120 °C – 180 °C) and different times (5 min – 75 min) was applied. Results indicate that 180 °C is optimal temperature for isolation of keratin with subcritical water. Presence of keratin was confirmed using FTIR analysis. Molecular weight of subcritical water isolation product was determined using SDS – PAGE electrophoresis and Size exclusion gel chromatography. The molecular weight of the keratin produced by the isolation reaction ranges between 4 kDa and 12 kDa. In conclusion, subcritical water acts as a reducing agent at higher temperatures, breaking the disulfide bond in the keratin molecule, which consequently decays into smaller units. Further research is needed to determine the viability of the isolation method from poultry feathers with subcritical water on an industrial scale.
