Title:Esterifikacija maščobnih kislin z biokatalizatorjem vezanim na mezoporozni silikatni nosilec : magistrsko delo

Abstract:Reakcija spajanja maščobnih kislin in alkohola, pri kateri nastane produkt ester, ima pomembno vlogo v vsakdanjem življenju in prav tako v kemijski industriji. Produkt, alkilni ester, je najpogostejša vrsta biodizla. V tej raziskavi smo sintetizirali in karakterizirali biokatalizator, pri katerem smo kot nosilec uporabili mezoporozni silicijev dioksid (SBA-15) z veliko specifično površino in enakomerno velikostjo por. S postopkom sinteze smo vplivali na povečanje premera por SBA-15 z uporabo triizopropilbezena (TIPB). Kot ne-specifično povezovalno sredstvo za imobilizacijo encima na modificiran nosilec smo uporabili glutaraldehid. Učinkovitost imobilizacije lipaze na sintetizirani mezoporozni siliki smo preverili z reakcijo esterifikacije oleinske in miristinske kisline. Eksperimentalni del je potekal v avtomatiziranem reakcijskem sistemu. Preučevali smo vpliv ključnih procesnih parametrov temperature,  = (20, 25 in 30) °C , pH = (6, 7 in 8), množinskega razmerja kislina/alkohol in količine biokatalizatorja, na učinkovitost imobilizacije in esterifikacije.

Title:Esterification of fatty acid using biocatalyst attached to mesoporous silicate support
Abstract:The combination of fatty acid and alchohol forms a product ester. Esters plays a significant role in everyday life and also in the chemical industry. The product alkyl ester is the most common type of biodiesel. In this study we synthesized and characterized a biocatalyst using mesoporous silica (SBA-15) as a support. The high surface area of mesoporous silica SBA-15 has been synthesized successfully by using triblock copolymer (P123) as a direct structure agent and tetraethyl orthosilicate (TEOS) as a precursor. 1,3,5-Triisopropylbenzene (TIPB) were used in order to increase ultralarge pore. Glutaraldehyde was used as a non-specific cross-linking agent for fixing the lipases on mesoporous silica. Enzyme was covalently bonded on the support. The efficiency of bonding was tested by well-known esterification of oleic and myristic acid. All experiments were carried out in the automated reaction system. We studied the influence of key process parameters: temperature, ϑ = (20, 25 and 30) ° C, pH = (6, 7 and 8), acid/alcohol ratio and amount of biocatalyst on immobilization and esterification efficiency.
