Title:Analiza genetske raznolikosti akcesij malin (Rubus idaeus L.) iz genske banke Fakultete za kmetijstvo in biosistemske vede z uporabo mikrosatelitskih markerjev (SSR) : magistrsko delo

Abstract:Maline (Rubus idaeus L.) so priljubljena sadna vrsta, primerna za svežo uporabo in predelavo. Pridelava je v zadnjem desetletju močno narasla, kar je spodbudilo potrebo po razvoju sort, odpornih na bolezni in škodljivce ter prilagojenih specifičnim rastnim pogojem. Večina današnjih kultivarjev izvira iz majhnega števila starševskih rastlin, zato je ključno, da se za potrebe žlahtnjenja zagotovi gensko raznolik material. Pomembno vlogo pri tem imajo genske banke, katerih uspešnost temelji na ustrezni karakterizaciji in natančni identifikaciji akcesij ter preprečevanju podvajanj. Namen magistrskega dela je bil z uporabo mikrosatelitskih markerjev preveriti genetsko sorodnost med akcesijami malin, vključenimi v gensko banko Fakultete za kmetijstvo in biosistemske vede, ter oceniti uporabnost izbranih mikrosatelitskih lokusov za tovrstne analize. Na osnovi podatkov kapilarne elektroforeze smo z metodo »neighbor-joining« (NJ) izdelali dendrogram, ki je omogočil vizualizacijo genetskih odnosov med akcesijami. Rezultati so razkrili prisotnost večjega števila duplikatov, večinoma v skupinah po 2–3 identične akcesije, v enem primeru pa jih je bilo zaznanih celo 8. Kljub visoki informativnosti uporabljenih markerjev so naši rezultati omejeni zaradi majhnega števila analiziranih mikrosatelitskih lokusov (4), kar zmanjšuje natančnost zaključkov.

Title:Analysis of genetic diversity in raspberry (Rubus idaeus l.) accessions from the plant gene bank of the Faculty of Agriculture and Life Sciences using microsatellite (SSR) markers
Abstract:Raspberries (Rubus idaeus L.) are a popular fruit, as they can be used fresh or processed in a variety of products. The production of raspberries has seen a significant incline in the past decade. Therefore, there is an evergrowing need for hardy varieties, adapted to specific growing locations. Because of the origin of most modern varieties being from a small number of parent plants, it is important for genetically diverse breeding material to be made accesible. Gene banks play an important role in the preservation of genetic diversity. Accessions in gene banks must be correctly characterised, identified, marked, and be free of duplicates. The purpose of this master's thesis was the analysis of genetic relatedness among raspberry accessions stored at the Faculty of Agriculture and Life Sciences gene bank via the use of microsatellite markers. One of the goals was also to analyse the utility of the chosen microsatellite markers for future research. Data produced from a capillary electrophoresis was used to construct a dendrogram, using the Neighbor-Joining (NJ) method. The dendrogram made it possible to visualize genetic relationships between accessions. The results showed a great number of discovered duplicates, mainly in groups of 2 to 3 accessions, 8 in a single case. However, methodological limitations due to a small number of analysed microsatellite loci (4) prevented definitive conclusions.
