Title:Razvoj in optimizacija procesa sinteze furosemida : magistrsko delo

Abstract:Magistrsko delo obravnava razvoj in optimizacijo procesa sinteze farmacevtske učinkovine furosemid s pomočjo z bakrom katalizirane Ullmannove kondenzacije. Namen dela je razvoj alternativne, trajnostne sintezne poti, ki z uporabo bakrove katalize in globoko evtektičnih topil (DES) nadomešča ostre reakcijske pogoje in strupena topila tradicionalnih industrijskih postopkov. V okviru eksperimentalnega dela je preučen vpliv različnih reakcijskih medijev, baz in bakrovih katalizatorjev na učinkovitost kondenzacije med 2,4-dikloro-5-sulfamoilbenzojsko kislino in furfurilaminom. Za nadaljnje sistematično raziskovanje eksperimentalnega prostora je uporabljen statistični pristop načrtovanja eksperimentov (DoE). Z delnim faktorskim presejalnim načrtom smo določili temperaturo, volumen topila in količino nukleofila kot ključne procesne parametre, ki smo jih nato optimizirali z metodo odzivnih površin (RSM). Ustreznost razvitega protokola smo potrdili z izvedbo reakcije na povečani skali ter izolacijo končnega produkta do farmacevtske čistosti.

Title:Fusosemide synthesis process development and optimization
Abstract:This master's thesis explores the development and optimization of a synthetic process for the active pharmaceutical ingredient furosemide, utilizing a copper-catalyzed Ullmann condensation. The aim of the research was to develop an alternative, sustainable synthetic pathway that replaces the harsh conditions and toxic solvents of traditional industrial procedures by employing copper catalysis and deep eutectic solvents (DES). During the experimental phase, the influence of various reaction media, bases, and copper catalysts on the efficiency of the condensation between 2,4-dichloro-5-sulfamoylbenzoic acid and furfurylamine was investigated. To further systematically explore the experimental space, a statistical Design of Experiments (DoE) approach was applied. Using a fractional factorial screening design, temperature, solvent volume, and nucleophile amount were identified as key process parameters, which were subsequently optimized using Response Surface Methodology (RSM). The feasibility of the developed protocol was confirmed by performing the reaction on a larger scale and isolating the final product to pharmaceutical-grade purity.
