Title:Sinergistično delovanje selena in kadmija na rast ozkolistnega koščca Berula erecta Huds. v tkivni kulturi : magistrsko delo

Abstract:Ozkolistni koščec (Berula erecta) je makrofit iz družine kobulnic s sposobnostjo hitre rasti in razvoja ter privzema različnih elementov in je zato zanimiv za fiziološke raziskave rastlin. Nizke koncentracije selena (Se) imajo dokazano pozitivne učinke tudi na rastline, zato smo raziskali ali lahko Se zaščiti B. erecta pred kadmijevim (Cd) stresom. B. erecta smo gojili na MS gojišču in jih po 14 dneh zalili z raztopinami z različnimi koncentracijami in kombinacijami Se (0,1 ali 1 mg L-1) in Cd (0 ali 10 ali 50 mg L-1). Močan negativen vpliv Cd v primerjavi s kontrolo brez Cd smo opazili pri najvišji koncentraciji 50 mg L-1 po štirih tednih kulture. Tudi 10 mg L-1 Cd je povzročalo rastlinam blažji stres. To se je kazalo v zmanjšani sveži masi, višini, dolžini korenin, količini klorofilov in antocianov. Najbolj spodbujevalen učinek Se na vse parametre je imel dodatek višje 1 mg L-1 koncentracija Se v MS gojišče brez Cd po štirih tednih kulture. Tam, kjer pa sta bila dodana 10 mg L-1 ali 50 mg L-1 Cd v gojišče, se je izrazil spodbujevalni učinek nižje 0,1 mg L-1koncentracija Se po štirih tednih kulture. S tem smo dokazali, da Se do neke mere deluje spodbujevalno ob stresnih dejavnikih, ki jih povzroča Cd stres. Obenem pa smo dokazali tudi, da višje koncentracije Se nimajo nujno pozitivnega učinka na rastlino, ampak se le ta lahko z elementom zasiči.

Title:Synergistic effect of selenium and cadmium on the growth of lesser water-parsnip Berula erecta Huds. in tissue culture
Abstract:Lesser water-parsnip (Berula erecta) is a macrophyte belonging to the Apiaceae family, characterised by rapid growth and development and by the uptake of various elements, and is therefore of interest for plant physiological research. Low concentrations of selenium (Se) have been shown to have positive effects on plants; we therefore investigated whether Se can protect B. erecta from cadmium (Cd) stress. We cultivated B. erecta on MS medium and, after 14 days, treated the plants with solutions containing various concentrations and combinations of Se (0.1 or 1 mg L⁻¹) and Cd (0, 10 or 50 mg L⁻¹). A strong negative effect of Cd, compared with the Cd-free control, was observed at the highest concentration of 50 mg L⁻¹ after four weeks of cultivation. Even 10 mg L⁻¹ Cd caused milder stress to the plants. This was reflected in reduced fresh weight, plant height, root length, and chlorophyll and anthocyanin contents. The most stimulatory effect of Se on all parameters was observed when a higher concentration of 1 mg L⁻¹ Se was added to the MS medium without Cd after four weeks of cultivation. However, when 10 mg L⁻¹ or 50 mg L⁻¹ Cd was added to the medium, the stimulatory effect was observed at the lower concentration of 0.1 mg L⁻¹ Se after four weeks of cultivation. We have thus demonstrated that selenium exerts a stimulatory effect, to a certain extent, in response to stress factors caused by cadmium. At the same time, we have also shown that higher concentrations of selenium do not necessarily have a positive effect on the plant, as the plant may simply become saturated with the element
